氧化乐果对雄性小鼠精液质量及骨髓细胞微核的影响

【摘要】 目的研究低剂量重复染毒有机磷农药氧化乐果对雄性小鼠精液质量和骨髓细胞微核的影响。方法将55只昆明雄性小鼠随机分成5组,其中三个染毒组、一个阴性对照组和一个阳性对照组,染毒组分别以1.5、3.0、6.0mg/kg bw灌胃氧化乐果,阴性对照组灌胃生理盐水,采用等容量灌胃法(0.2mg/kg.bw),阳性对照组腹腔注射环磷酰胺(40mg/kg.bw),连续5天。染毒结束后通过精液质量分析和骨髓嗜多染红细胞微核试验来研究氧化乐果对雄性小鼠的生殖毒性和遗传毒性。结果随着氧化乐果染毒剂量的逐渐增加,小鼠肝脏重量、精子密度、存活率、活动度均明显下降,骨髓细胞微核数目显著增加,与对照组比较差异有统计学意义(P<0.05)。结论氧化乐果可引起小鼠精液质量下降和骨髓嗜多染红细胞遗传毒性。

【Abstract】 Objective To explore the Effect of Omethoate which repeated exposure of low doses on the Quality of Sperm and micronucleus of Bone-marrow Cell in Male Mice Methods 55 healthy male Kunming mice were randomly divided into five groups including three treatment groups,a negative control group and a cyclophosphamide group.The treatment groups were exposed respectively to omethoate at dosage of 1.5mg / kg(the low dose group),3.0 mg / kg(the moderate dose group) and 6.0 mg / kg(the high dose group).fed oxidation Music results,the negative control group was exposed 0.9%saline with Equivalent capacity of 0.2mg/kg.The cyclophosphamide group was exposed with intraperitoneal injection of 40mg/kg.bw.Whole mice were raised for 5 days.Results As the omethoate doses was increased,the liver weight,the density of sperm,the rate of living sperm were decreased significantly and the number of micronucleus in Bone-marrow cell were increased evidently compared with the control group(P<0.05).The testis weight and epididymal viscera of mice were not statistically significant compared with the control group(P>0.05).Conclusion Omethoate can decrease the quality of sperm in mice and cause Genetic toxicity in bone-marrow polychromatic erythrocytes.
