福建水华微囊藻毒素在体内外对免疫功能的影响

【摘要】 目的探讨福建水华微囊藻毒素免疫毒性。方法 80只雄性ICR小鼠随机分成四组,微囊藻毒素染毒剂量分别为0,8,16,24μg/kg,连续7d,第8天检测脾脏的超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH-PX)、丙二醛(MDA)、乳酸脱氢酶(LDH)、巨噬细胞吞噬指数、T淋巴细胞CD4~+、CD8~+亚群的百分比,观察胸腺和脾脏的组织病理学改变;无菌取小鼠脾细胞和腹腔巨噬细胞,MC-LR染毒剂量均为0、1、5、10μg/ml,观察MC-LR对小鼠T、B淋巴细胞增殖的影响和MC-LR染毒24、48和72 h对小鼠巨噬细胞增殖的影响。结果 24μg/kg微囊藻毒素可使SOD、GSH-PX活性明显降低,LDH水平明显升高,吞噬指数明显下降,CD4~+的百分比明显降低(P<0.05),胸腺皮质明显变薄,脾小体明显萎缩,淋巴细胞数明显减少;1、5、10μg/ml剂量组T淋巴细胞刺激指数(SI)下降P<0.05),5、10μg/ml剂量组B淋巴细胞刺激指数(SI)下降(P<0.05),染毒48 h各剂量组腹腔巨噬细胞增殖受到不同程度抑制,其中5、10μg/ml剂量组抑制增殖明显(P<0.05),染毒72 h 1、5、10μg/ml剂量组抑制增殖明显(P<0.05),呈现一定的时间和剂量关系。结论福建水华微囊藻毒素具有免疫毒性,需进一步加强对饮用水微囊藻毒素污染的防控。

【Abstract】 Objective To study immunotoxicity and its mechanism of microcystins from cyanobacteria blooms in mice in Fujian.Methods:80 ICR male mice were assigned to four groups and treated with different dose of 0,8,16,24μg/kg microcystins for 7 days.On the eighth day,SOD,GSH-PX,MDA,LDH, phagocytic index of macrophage,the percentage of CD4+ and CD8+ were tested,then thymus and spleen was examined with HE staining.Mice splenocytes were treated with MC-LR at the doses of 0,1,5,10μg /ml,then tested for proliferation of T lymphocytes and B lymphocytes;Mice peritoneal macrophages were treated with MC-LR at the doses of 0,1,5,10μg /ml for 24,48,72h,then its proliferation was tested Results SOD,GSH-PX,phagocytic index of macrophage,the percentage of CD4+ were all decreased and LDH was increased significantly in group treated with 24μg/kg compared with the control group(P<0.05).Meanwhile,The depth of thymic cortex, the spleen lobule,the numbers of lymphocyte were reduced markedly.The proliferation of T lymphocytes was decreased at the doses of 1,5,10μg /ml(P<0.05),and the proliferation of B lymphocytes was decreased at the doses of 5,10μg/ml(P<0.05).The proliferation of peritoneal macrophages was suppressed at the doses of 5,10μg /ml after 48h and at the dose of 1,5,10μg /ml after72h(P<0.05).The proliferation was reduced in a dose and time-dependent manner.Conclusion In this study the microcystins from cyanobacteria bloom in Fujian had immunotoxicity.Microcystins should be prevented and control in order to improve drinking water sanitation
