氯化镍亚慢性染毒大鼠肺组织脂质过氧化与抑癌基因表达的相关性研究

【摘要】 目的探讨不同浓度氯化镍染毒大鼠肺组织脂质过氧化和抑癌基因p15、p16表达水平以及二者之间的相关性。方法以0、1、2、4和8mg/kg体重灌胃染毒大鼠,持续6周。染毒结束后处死大鼠,制备肺组织匀浆,采用分光光度法,测定超氧化物歧化酶(SOD)和丙二醛(MDA)水平的变化;以荧光定量PCR法检测肺组织中抑癌基因p15和p16的表达水平。结果随着氯化镍染毒剂量的增加,SOD活性下降,MDA含量增多,且各染毒组与对照组相比差异有统计学意义(P<0.01)。除1mg/kg体重组p16mRNA表达量与对照组相比无统计学差异外,其他各组p15和p16mRNA表达量与对照组相比均显著性降低(P<0.01)。经pearson相关分析,p15、p16与SOD表达显著性正相关,而与MDA则为显著负相关(P<0.05)。结论氯化镍亚慢性染毒大鼠肺组织脂质过氧化水平与抑癌基因p15和p16表达具有一定的相关性。

【Abstract】 Objectives To investigate the dynamic changes and correlations of Lipid Peroxidation with anti-oncogene in Lung of Rat exposed to nickel chloride.Methods The rats were exposed to nickel chloride at concentrations of 0,1,2,4 and 8mg/kg b.w.by intragastric administration.All groups were administrated 5 times a week for six weeks.The levels of malonydialdehyde(MDA) and superoxide dismutase(SOD) in the lung of rats were measured.The expressions of anti-oncogene of p15 and p16 were detected by real time polymerase chain reaction.Results Compared with the control group,the levels of MDA of lung in the rats exposed to nickel chloride increased significantly,while the levels of SOD decreased significantly(P<0.01).The expression of p15 decreased significantly as the concentrations of nickel chloride increased(P<0.01),the levels of p16 in the groups at concentrations of nickel chloride>2mg/kg b.w.significantly decreased compared with the control(P<0.01).In addition,there was a significant positive correlation between anti-oncogene and SOD,and a negative correlation was founded between anti-oncogene and MDA.Conclusion These results suggested that a close correlation exists between lipid peroxidation and the expressions of p15 and p16 in lung of rats exposed to nickel chloride.However,the mechanisms underlying this observation need further investigation.
