玉米×四倍体多年生玉米杂种F2非整倍体的研究

【摘要】 用遮光调节开花时期、喷施低浓度赤霉素等提高可杂交性措施、利用玉米与四倍体多年生玉米杂种F1合子发育过程中染色体配对不正常发生染色体消失得到少量非整倍体F2。对获得的几个F2植株进行形态观察、细胞学和SSR鉴定。结果表明,10株F2植株形态特征变化较大,有6株野生亲本型植株,2株中间型植株,2株玉米型植株;它们的体细胞数目为24、25、26,其中25条的频率较高;SSR标记鉴定结果表明,玉米的第2,3,4,5染色体较1,7,8,9,10染色体易于在F2中消除,间接证明玉米×四倍体多年生玉米杂种F1(2n=30)减数分裂单价体为玉米的第2,3,4,5染色体可能性大于玉米的其它染色体。另外,还对利用玉米与四倍体多年生玉米非整倍体染色体消除现象产生的原因和玉米与四倍体多年生玉米非整倍体染色体在玉米遗传育种研究的应用进行了讨论。
【Abstract】 Although the F1 of maize×Z.perennis has genomic affinities,few F2 generation could be obtained by using some specially treatment.In this study,morphologic,cytological and SSR observation on F2 were conducted.Results showed that 10 F2 plants have different plant types such as wild parent-like,internal type,maize-like,these F2 cells have 24,25 and 26 chromosomes frequency,which were aneuploids.Duo to maize×Z.perennis(2n=30) plant with the most frequent configuration of 5Ⅲ+5Ⅱ+5Ⅰ,which produced the chromosome elimination for aneuploids.The SSR assay further confirmed that maize chromosome 2,3,4,5 were easy to be eliminated than maize chromosome 1,7,8,9,10,which could be deduced that maize chromosome 2,3,4,5 were easy to be univalents in meiotic configuration of F1 hybrid of maize×Z perennis.Among these F2 aneuploids,it can be produced as addition or substitute line by cytogenesis project.
