乙酰化改性蚕蛹蛋白条件的优化

【摘要】 本文采用乙酸酐对蚕蛹蛋白进行酰化改性。在进行了蛋白浓度、乙酸酐/蛋白量和反应时间对蚕蛹蛋白的乙酰化程度影响的单因素试验基础上,以乙酰化程度为评价指标,通过响应面分析法对乙酰化改性蚕蛹蛋白的条件进行了优化,并对改性后蚕蛹蛋白的溶解度进行了研究。结果表明,最佳工艺条件为:蛋白浓度6.0%、乙酸酐/蛋白量22.0%、反应时间59.8min。理论最大乙酰化程度值为83.20%,验证值为82.82±1.49%,说明预测模型可靠性高,可用于反应条件的优化。此外,溶解度试验结果表明,改性后的蚕蛹蛋白溶解度显著提高,比改性前提高了39.27%。
【Abstract】 In this study,acetic anhydride was used to modify the silkworm pupae protein. Effects of protein concentration,the ratio of acetic anhydride to protein and reaction time on acetylation of silkworm pupae protein were studied by single factor experiment.On the basis of these,response surface analytical method,in which acetylation was taken as response value,was used to optimize the conditions of acetylation modification for silkworm pupae protein.The effect of acetylation modification on the solubility of silkworm pupae protein was also investigated.The results showed that the optimal processing parameters were as follows:protein concentration 6.0 %,the ratio of acetic anhydride to protein 22.0%and reaction time 59.8min.Through the established mathematical model,the highest acetylation value was 83.20%in theory and the actual value was 82.82±1.49%.The result showed that the established mathematical model is reliable.In addtion,the result of solubility experiment showed that the solubility of silkworm pupae protein after acetylation modification was remarkably improved,and its solubility increased by 39.27%.
