桑树内生真菌Macrophomina phaseolina MOD-1生产油脂及降解纤维素性能研究

【摘要】 从桑树中分离获得一株产油内生真菌Macrophomina phaseolina MOD-1,用苏丹Ⅲ染色法发现此菌株菌丝及菌核内均可产生大量油脂类物质。用酸热法提取油脂,油脂甲酯化后采用气-质联用仪进行脂肪酸组成分析,发现该菌株油脂含量为细胞干重的22.72%,包含18种脂肪酸,其中不饱和脂肪酸含量为69.88%,主要为反油酸(37.80%)、亚油酸(13.55%)、棕榈油酸(10.71%)、油酸(6.72%),还有少量的十四碳烯酸、二十碳二烯酸、二十碳四烯酸等不饱和脂肪酸;饱和脂肪酸含量为30.12%,主要为棕榈酸(19.56%)、硬脂酸(8.22%)、花生酸(1.11%)、肉豆蔻酸(0.51%)以及少量的月桂酸、十五烷酸、十七烷酸、山嵛酸等饱和脂肪酸。同时,该菌株能在以羧甲基纤维素钠为唯一碳源的培养基中生长,其发酵液中滤纸酶活为88.80U,C1酶活为165.24U,Cx酶活为332.27U。本研究为利用微生物转化秸秆生产油脂奠定了坚实的基础。
【Abstract】 Endophytic fungus Macrophomina phaseolina MOD-1 strain was isolated from the root of mulberry.Its mycelia and sclerotia produced large amount of the substances of oil kinds that was stained with Sudan Ⅲ.The extracted oils by thermal acid method was methyl esterified,18 kinds of fatty acids were detected out according qualitative analysis and relative content analysis of fatty acids by GC/MS.The content of unsaturated fatty acids was 69.88%,mainly included elaidic acid (37.80%),linoleic acid (13.55%),palmitoleic acid (10.71%),oleic acid (6.72%),and also a little tetradecanoic acid,eicosadienoic acid,arachidonic acid etc;while the content of saturated fatty acids was 30.12%,mainly included palmitic acid (19.56%),octadecanoic acid (8.22%),eicosanoic acid (1.11%),tetradecanoic acid (0.51%) and also a little dodecanoic acid,pentadecanoic acid,docosanoic acid etc.The strain could accumulate lipid and they had potential ability of assimilating cellulose.This research is the foundation for applying those strains which can accumulate lipid and simultaneously assimilate the cellulose in straw.
