高温干燥对家蚕胚胎发育影响的蛋白质表达谱分析

【摘要】 采用双向电泳和图像分析技术,对家蚕温敏性品种"限伴"在常温常湿和高温干燥催青条件下转青期胚胎的蛋白表达变化进行了研究。发现在不同催青条件下的蚕胚胎蛋白成分在质和量上都有明显的差异。在易溶蛋白组分中,常温常湿催青处理区和高温干燥催青处理区分别有67个和76个特异蛋白点,分别占各自总蛋白点数的15.95%和17.72%,匹配的蛋白点,表达量差异超过3倍以上的分别有18个和35个。在难溶性蛋白组分中,常温常湿催青处理区和高温干燥催青处理区的特异蛋白点则为35个和33个,分别占总蛋白点数的15.42%和14.67%,能匹配且表达量差异超过3倍以上的蛋白点分别有19个和30个。比对高温干燥催青处理区与常温常湿催青处理区的特异表达蛋白,点数差异不大,但可匹配的蛋白点,在表达量方面,有明显的增多。推测在高温干燥催青条件下导致"限伴"雄性不孵化的原因,除了因高温干燥诱导或关闭某些基因表达外,还可能与某些基因的超量表达或受抑制有关。

【Abstract】 The changes of protein expression of embryo at blue egg stage were compared using dielectrophoresis (2-DE) and image analysis under incubation condition of normal temperature and humidity or high temperature and dry condition from temperature sensitive Silkworm of "Xianban".The results showed that the protein expression of embryo appeared significant difference in protein composition and expression level under different incubation condition.In the soluble protein,there is 67 and 76 specific protein in silkworm embryo incubated under normal temperature and humidity or high temperature and dry condition,and share 15.95% and 17.72% of their total protein,respectively.The proteins expressed over 3 fold level are 18 in normal temperature and humidity incubation and 35 in high temperature and dry condition incubation among matched proteins.In the insoluble protein,there is 35 and 33 specific protein in silkworm embryo incubated under normal temperature and humidity or high temperature and dry condition,and occupy 15.95% and 17.72% of their total protein,respectively.The proteins expressed over 3 fold level are 19 in normal temperature and humidity incubation and 30 in high temperature and dry condition incubation among matched proteins.Comparing the changes of protein expression between silkworm embryos incubated under normal temperature and humidity and high temperature and dry condition,the amount of specific proteins is similar,but the amount of proteins expressed over 3 fold in matched proteins increase in the silkworm embryo incubated under high temperature and dry condition.It is suggesting that unhatched male incubated under high temperature and dry condition may involve in some genes over-expressed or suppressed,besides some genes are induced or silenced.
