茶树密码子用法分析

【摘要】 本研究运用CodonW软件和CUSP程序对筛选的134个茶树蛋白质编码基因序列进行了分析、计算了密码子使用频率,并将其与人、果蝇、酵母、大肠杆菌这4种模式生物及拟南芥、大豆、棉花、水稻、小麦等单、双子叶植物进行比较。结果显示茶树密码子偏爱性与人、果蝇、酵母、大肠杆菌有不同程度的差异,与水稻、小麦单子叶植物的密码子使用频率差异较大,与拟南芥、大豆、棉花双子叶植物的密码子偏爱性一致,分析结果对茶树基因转化及高效表达系统的选择等具有重要指导意义。

【Abstract】 In this study,coding sequence of 134 selected proteins were analyzed by CodonW and CUSP programs in Camellia sinensis,and we also calculating frequency of codon usage which encoded amino acids and comparing with variety classes of mode organisms such as:Homo sapiens,Saccharomyces cerevisiae,Escherichia coli,Drosophila melanogaster.These results had shown that codon bias of tea plant was difference in various degrees with these mode organisms. Comparing codon usage frequency of tea plant with dicotyledons and monocotyledons including Arabidopsis thaliana,Glycine max,Gossypium hirsutum,Oryza sative,Triticum aestivum,the consequence indicated that codon usage frequency of tea plant was obviously different from Oryza sative and Triticum aestivum which belonged to monocotyledons,but had a consistency with codon bias of Arabidopsis thaliana,Glycine max and Gossypium hirsutum which are dicotyledons.These analytic results had important guidance for tea plant gene-transforming and selecting the high-efficient expression system.
