应用RAPD和EST-SSR分析铁观音与黄棪杂交一代的遗传差异
Analysis of Genetic Variance of Tie Guan Yin and Huang Dan Hybrid Generration Using RAPD and EST-SSR Markers

【摘要】 应用EST-SSR和RAPD标记对铁观音与黄棪杂交一代进行遗传多样性和亲缘关系分析。25条RAPD引物共扩增出181个等位位点,平均7.24个,PIC平均0.756,Shannon信息指数(Ⅰ)平均0.331,Nei’s基因多样性(h)平均0.215,UPGMA聚类分析,在相似系数0.84处可将17个参试材料分为5类,应用主坐标分析,亦可分为5类。24对EST-SSR引物共扩增出99个等位位点,平均4.13个,PIC平均0.561,Shannon信息指数(Ⅰ)平均0.309,Nei’s基因多样性(h)平均0.199,UPGMA聚类分析,在相似系数0.84处可将17个参试材料分为5类,应用主坐标分析,亦可分为5类。结果看出,铁观音与黄棱杂交一代品种间存在一定的遗传差异。两种标记分析的结果不完全一致,但是,均显示出杂交一代与两亲的亲缘关系较密切。说明高香优质品种铁观音与黄棱的杂交一代具有两亲本厚实的遗传基础。黄观音、金观音与父本黄棪,金牡丹、金桂观音与母本铁观音的亲缘关系相对更近。铁观音、黄棱及其杂交一代与参照品种白奇观音、大红袍、苦味白牡丹、迎春、台茶12号之间的亲缘关系相对较远。
【Abstract】 The EST-SSR and RAPD were used to analyze the genetic polymorphism and genetic relationship of hybrids between Tie guan yin and Huang dan.25 RAPD primers amplified 181alleles,with an average of 7.24.The average value of primer polymorphism information content (PIC),Shannon information index(I) and Nei’s gene diversity(h) was 0.756,0.331 and 0.215 respectively.17 tested materials were divided into 5 categories at a relation coefficient of 0.84 by UPGMA cluster analysis method and principal coordinates analysis method.24 pairs of EST-SSR primers amplified 99 alleles,with an average of 4.13.The average value of primer polymorphism information content(PIC),Shannon information index(I) and Nei’s gene diversity(h) was 0.561, 0.309 and 0.199 respectively.17 tested materials were also divided into 5 categories at a relation coefficient of 0.84 by UPGMA cluster analysis method and principal coordinates analysis method. Results showed that there were some genetic differences among different varieties from the hybrids between Tie Guan Yin and Huang Dan.Analysis results of the two markers are not totally consistent,but all showed closer genetic relationship between the hybrids and their parents,which suggesting there was strong genetic basis between the hybrids and their parents of Tie Guan Yin and Huang Dan which had high incense and better quality.There was close relationship between the male parent Huang Dan and its hybrids of Huang Guan Yin and Jin Guan Yin,so did the female parent Tie Guan Yin and its hybrids of Jin Mu Dan and Jin Gui Guan Yin.Tie Guan Yin and Huang Dan together with their hybrids had distant genetic relationship with reference varieties Bai Qi Guan Yin,Da Hong Pao,Ku Wei Bai Mu Dan,Ying Chun and Tai Cha12.
