【摘要】 利用ISSR分子标记技术对黄金茶珍稀地方种质资源的110个株系进行了遗传多样性和亲缘关系分析。从100条ISSR引物中筛选出18条多态性高、稳定性强的引物分别对供试材料的基因组进行扩增,共获得205条清晰可辨的谱带,其中多态性带187条,多态性条带比率为91.22%,这表明供试材料的DNA多态性程度较高。Popgen1.32软件统计结果显示,110个黄金茶株系平均有效等位基因数为1.5092,平均Nei’s基因多样性指数为0.3014,平均Shannon信息指数为0.4547,说明该群体的遗传多样性水平较高。应用NTSYSpc-2.10e软件计算得到各株系间的Jaccard相似系数介于0.22～1.00之间,平均相似系数为0.55。根据遗传相似系数,用UPGMA法将黄金茶群体110个株系分成七大类群,并分析了各株系间的亲缘关系。更多还原

【Abstract】 Genetic diversity and relationship of 110 "Huangjincha"〔C. sinensis (L.) O. Kuntze〕strains were analyzed by inter-simple sequence repeat (ISSR) molecular markers. A total of 205 bands were detected by using 18 ISSR primers, of which 187 were polymorphic, and the total percentage of polymorphic bands was 91.22%, the result showed that abundantdiversityexisted among tea strains studied. As analyzed by POPGENE 1.32, the average value of effective number of alleles, Nei’s gene diversity and Shannon’s information index were 1.5092, 0.3014 and 0.4547 respectively, which indicated that the genetic diversity of Huangjincha population was relatively high. As analyzed by NTSYSpc-2.10e, the Jaccard similarity coefficient of 110 tea strains ranged from 0.22 to 1.00, and the average was 0.55. The 110 tea strains were divided into seven groups by UPGMA (unweighted pair group method with arithmeticaverage) , and the genetic relationships of them were analyzed according to the similarity coefficient.更多还原
