鼻咽癌高癌家系成员基因组稳定与修复基因表达研究

【摘要】 目的:研究鼻咽癌高癌家系成员基因组稳定与修复基因的表达情况。方法:在鼻咽癌高发区广东中山市,选择鼻咽癌高癌家系成员为研究对象,以鼻咽癌家系成员、鼻咽炎、鼻咽癌前病变及鼻咽癌患者为对照,从基因组稳定和修复功能分类基因角度,以芯片技术进行研究。结果:鼻咽癌高癌家系成员DNA损伤修复基因主要以上调表达为主,且上调幅度高,上调100倍以上的基因包括:TEP1、MLH3、MSH4、PMS2L1、OGG1;下调的基因少且幅度低,泛素类基因多表现为下调。结论:鼻咽癌高癌家系成员存在着特定的DNA损伤修复基因变化规律。

【Abstract】 Objective To observe the expression of genes related to genome stability and DNA repair in the members of Nasopharyngeal Carcinoma(NPC) clustering families.Methods In the Zhongshan city which there is highly incidence rate of NPC,we chose the members of the NPC clustering families as object,and the member of the NPC family,nasopharyngitis,nasopharyngeal precancerous lesion and NPC as the control group.The microarray-based technique was used to examine the genome stability and DNA repair genes in the nasopharyngeal tissue.Results More Genome Stability and DNA Repair Genes were up-regulated in the members of the NPC clustering families,and the range of up-regulated is high,the over up-regulated 100 times genes including TEP1、MLH3、MSH4、PMS2L1、OGG1.Few Genome Stability and DNA Repair Genes were down-regulated in the members of the NPC clustering families,the ubiquitin genes almost were down-regulated.Conclusion There are specially expression character of genome stability and DNA repair genes in the the members of NPC clustering families.
