水稻苯达松敏感型突变体嘉浙mA/mB的选育

【摘要】 以剂量为350Gy的γ射线辐射处理"三系"保持系嘉浙B水稻种子,经M2代筛选和M3、M4代验证,获得一份对苯达松高度敏感的突变体嘉浙mB。在幼苗期喷施不同浓度的苯达松溶液,发现0.06%的苯达松即可使嘉浙mB致死;在成株期,叶面喷施0.5%的苯达松溶液也具致死效果。用嘉浙B/嘉浙mB杂交后代所做的遗传分析表明,嘉浙mB的苯达松敏感性状由隐性单基因控制。该突变基因与已报道的M8077S中的苯达松敏感基因等位,对已知控制苯达松敏感特性、位于第3染色体上的一个细胞色素P450基因(CYP81A6)的所做的分析表明,嘉浙mB与嘉浙B相比,该基因第一外显子第413位缺失了一个碱基T,造成移码突变,造成翻译提前终止,使该蛋白从513aa截短为175aa,使其失去活性。
【Abstract】 Seeds of cytoplasmic male sterile maintainer line JiazheB were irradiated with 60Co γ-rays at the dose of 350Gy.One mutant sensitive to the herbicide bentazon was selected in M2 generation selection, and was further verified in M3and M4 generation. Treatment of bentazon solutions of different concentrations showed that the lethal concentration was about 0.06% and 0.5% at seedling stage and at adult period, respectively. Genetic analyses of the segregating population derived from the cross between JiazheB and Jiazhe mB demonstrated that the bentazon-sensitivity trait is controlled by a single recessive nuclear gene. Allelism test showed that the mutant gene of Jiazhe mB is allelic to that of M8077S, which was reported as one variety sensitive to bentazon. According to a previous study, the betazon susceptibility is controlled by a P450 gene (CYP81A6). Sequence comparision of the CYP81A6 gene of JiazheB and Jiazhe mB showed that a single -base deletion of T at the 413th nueleotide in the first exon in Jiazhe mB, which might have caused a reading-frame shift and led to the premature termination of translation and a truncated protein of 175 aa as compared with 513 aa in wild type.
