肠易激综合征大鼠模型的复制与评价

【摘要】 目的建立肠易激综合征(IBS)大鼠模型,为实验药理学提供方法。方法采用复合因素诱导大鼠建立IBS模型,随机分为正常组、模型组和阳性组(匹维溴铵15.0mg/kg),每组10只,连续给药31d,分别于造模后和给药第10d、20d、30d测定各组大鼠的体重、摄食量、排便情况、自主运动量,末次给药60min后测定胃排空率和肠推进率,放射免疫法测定血清5-HT、血浆SP、VIP含量以及结肠匀浆5-HT、SP、VIP含量。结果造模后各大鼠体重减轻、摄食量减少,排便量增多、出现稀便和无定形软便,自主运动量减少,胃排空率减小、肠推进率加快,血清5-HT含量升高、血浆SP和VIP含量均降低,结肠匀浆5-HT和SP、VIP含量升高(P<0.05,P<0.01);给药第10d,阳性组大鼠排便量减少、稀便减少(P<0.05)。匹维溴铵具有一定的治疗作用,促使其向正常水平恢复。结论复合因素随机刺激能成功复制IBS大鼠模型,其病理生理特征与临床研究结果基本吻合。

【Abstract】 AIM To establish the Irritable bowel syndrome model in rats.METHODS SD rats were divided into the normal control group,model group,positive control group(Pinaverium Bromide Tablets 15.0mg/kg),the treatment course lasted 31 days.Body weights,appetite,defecation,independent exercise of all rats were detected.The rate of gastric emptying and small intestinal propulsion rate were measured.The 5-HT in serum,SP and VIP in plasma or 5-HT,SP,VIP in colon homogenates were evaluated by radioimmunoassay.RESULTS After modeling,the rats weight and food intake were decreased,stool quantity was increased,The independent movement was reduced,gastric emptying rates were decreased,intestinal propulsion rates were increase,the content of SP and VIP in blood were decreased,but increased in colonic homogenate(P<0.05,P<0.01).After treatment,Stool quantity was decreased,the food intake was increased and stool quantity was decreased,the body weight of rats were significantly increased,the amount of exercise and stool returned to normal,the 5-HT levels in serum or in colonic homogenate were significantly decreased,but VIP levels in plasma were markedly increased,the SP and VIP contents were significantly decreased in colonic homogenate in positive control group(P<0.05,P<0.01).CONCLUSION The stimulation of composite factors can successfully reproduce the rat model of IBS.
