黄芪甲苷对烧烫伤皮肤修复的研究

【摘要】 目的:建立稳定、规范的烧烫伤创面模型,研究黄芪甲苷溶液对烧烫伤创面修复的药理活性。方法:应用恒温恒压烫伤仪建立SD大鼠深二度烧烫伤创面模型,采用马森三染色法考察黄芪甲苷溶液在不同浓度、不同给药频率下对于烧烫伤创面修复的能力,并采用扫描电镜法观查皮肤内层胶原蛋白排列。结果:采用恒温恒压烫伤仪,烫头面积4cm2、烫头温度80℃,致伤压力为烫伤棒自重0.5kg,烫伤时间为8s可成功建立稳定、规范的SD大鼠深二度烧烫伤创面模型。在体内烧烫伤创面治疗实验中,黄芪甲苷溶液能加快创面愈合,促进皮肤血管的生成,规则胶原的有序排列。其治疗效果与给药浓度及给药频率成正相关。结论:黄芪甲苷在烧烫伤治疗中有很好的应用前景,后续以药剂学手段进一步提高治疗效果、延长给药间隔更具有重要意义。
【Abstract】 Objective: To establish a stabilized burn/scald rat model for drug evaluation, research the pharmacological activity of Astragaloside IV in wound healing of burn/scald.Methods: To establish the deep partial-thickness scald wound with SD rats by the instrument with constant temperature and pressure. Research the burn/scald wound repair capacity of Astragaloside IV under different concentration and frequency with masson’s trichrome stain analysis, and observe the collagen deposition in the intimal skin with scanning electron microscopy (SEM). Results: Scald for 8s was indentified as injury time for deep partial-thickness scald wound by the instrument with constant temperature(80℃) and pressure(0.5kg).In vivo, masson’s trichrome stain analysis indicated that Astragaloside IV may improve the wound healing and promoted the angiogenesis and collagen synthesis. The treatment effect of Astragaloside IV is concentration dependent and is also related with the dosing interval time. Conclusions: Astragaloside IV was shown a promising natural product with healing effect for burn/scald wound treatment. It is significant to improve the bioavailability of Astragaloside IV with the method of pharmaceutics in the treatment of burn/scald.
