海绵共附生疣孢菌FIM06054细胞毒活性次级代谢产物的研究

【摘要】 利用正相硅胶、葡聚糖凝胶Sephadex LH-20和反相C18柱层析等方法,从海绵共附生疣孢菌FIM06054的发酵代谢产物提取液中分离到二个化合物(1-2)。通过ESI-MS、NMR和UV等波谱方法鉴定其中一个化合物06054-1(1)与抗肿瘤化合物Proximicin A同质。另一个化合物06054-2(2)为新结构化合物。活性研究表明06054-1对肿瘤细胞MDA-MB-23l、SW620、SMMC7721和CNE-2具增殖抑制活性,其IC50值(μM)分别为23.49、13.48、34.96和16.28。06054-2对肿瘤细胞CNE-2具显著增殖抑制活性,其IC50值为6.88μM,但对肿瘤细胞株MDA-MB-231、SW620和SMMC7721的增殖抑制作用不明显。
【Abstract】 Two compounds(1-2) were isolated from culture broth of Verrucosispora sp.FIM06054 from marine sponge by silica gel,sephadex LH-20 and C18 reversed phase column chromatography.On the basis of spectral data,they were identified as antitumor compound Proximicin A(1) and a novel compound 06054-2(2).Compound 1 showed growth inhibitory activity against the tumor cell lines MDA-MB-231、SW620、SMMC7721 and CNE-2 with IC50 values of 23.49μM、13.48μM、34.96μM and 16.28μM respectivity in vitro by MTT assay,while Compound 2 showed prominent growth inhibitory activity against CNE-2 cells with IC50 values of 6.88μM.
