退银汤治疗银屑病动物模型的实验研究

【摘要】 目的研究退银汤对银屑病动物模型的治疗作用。方法将小鼠或豚鼠随机分为空白对照组、模型对照组、阳性药(消银颗粒)对照组,退银汤低、中、高剂量组,每组10只,各用药组灌胃给药,1次·d-1,连续14d,空白对照组和模型对照组给予等容量的生理盐水。采用雌激素期小鼠阴道上皮和小鼠尾部鳞片表皮病理模型,观察退银汤对雌激素期小鼠阴道上皮细胞有丝分裂指数及小鼠尾部表皮鳞片中有颗粒层鳞片数的影响;采用豚鼠耳廓皮肤银屑样皮损模型,观察皮损组织病理变化及耳廓表皮厚度;采用ELISA试剂盒检测豚鼠耳廓皮肤匀浆液中肿瘤坏死因子(TNF-α)及细胞间黏附因子(ICAM-1)的含量。结果退银汤能明显抑制雌激素期小鼠阴道上皮细胞有丝分裂,对小鼠尾部鳞片表皮颗粒层形成有显著促进作用,使豚鼠银屑病样模型耳廓皮肤厚度显著减少,豚鼠耳廓皮肤匀浆液中TNF-α及ICAM-1含量显著降低,并均呈良好的量效依赖关系。结论退银汤对银屑病动物模型有良好的治疗作用,为临床应用提供了实验依据。

【Abstract】 Objective To study treatment effect of Anti-Psoriasis Decoction on psoriasis animal models.Methods Rats or guinea pigs are randomly divided into blank control group,model control group,positive drug ( Anti-Psoriasis particles ) control group,low,medium and high dose groups of Anti-Psoriasis Decoction with 10 rats in each group.Treating groups are given Intragastric administration,once a day with continuously 14 days.Blank control group and model control group are given the same volumes of physiological saline.Use pathological models of mouse vaginal epithelium within estrogen period and scale epidermis of mouse tail to observe the influence of Anti-Psoriasis Decoction on mitosis index of mouse vaginal epithelium and scale amount in the granular layer of mouse tail epidermis.Use psoriasis-like guinea pig ear skin lesions model to observe pathological changes and thickness of ear epidermal,and use ELISA kit to detect the contents of tumor necrosis factor ( TNF-α) and intercellular adhesion molecule ( ICAM-1)in guinea pig ear skin homogenate.Results Anti-Psoriasis Decoction can obviously inhibit mitosis of mouse vaginal epithelium in estrogen period,and have distinct stimulative effect on the formation of scale epidermis granular layer of mouse tail,and the skin thickness of psoriasis-like model of guinea pig ear decreases significantly.TNF-A and ICAM-1 content in guinea pig ear skin homogenate reduce significantly,and show a good dose-effect relationship.Conclusions Anti-Psoriasis Decoction has good effect on treating psoriasis animal models,providing the experimental basis for clinical application.
