UPLC法同时测定脑心通胶囊中5个成分的含量

【摘要】 目的:建立超高效液相色谱(UPLC)同时测定脑心通胶囊中羟基红花黄色素A、阿魏酸、丹酚酸B、藁本内酯5个主要化学成分的方法。方法 :采用UPLC色谱系统,BEH C18色谱柱(2.1mm×100mm,1.7μm);流动相为0.5%甲酸-乙腈梯度洗脱;流速为0.3mL·min-1;检测波长羟基红花黄色素A为400nm,芍药苷为235nm,丹酚酸B为280nm,阿魏酸和藁本内酯为324nm。结果 :本方法可在24min内完成一次色谱分析,各主要成分色谱峰之间有良好的分离度,5个成分的浓度和各自峰面积之间有着良好的线性关系,精密度、重复性及加样回收率的RSD均小于2.0%。结论 :本方法快捷、准确、重复性好,能同时测定脑心通胶囊中5个主要化学成分,可较全面地控制脑心通胶囊这些化学成分的含量。
【Abstract】 Objective: To establish a UPLC method for simultaneous determination of five compounds (Hydroxysafflor yellow A ,Paeoniflorin,Ferulic acid,Salvianolic acid B,Ligustilide) in Naoxintong capsule.Methods: Five compounds in Naoxintong capsule were simultaneously determined by UPLC with WATERS ACQUITY UPLC BEH C18(2.1 mm ×100 mm,1.7 μm) column.The mobile phase consisted of 0.5% methanoic acid solution- acetonitrile with a gradient elution program,at the flow rate of 0.3mL·min-1,the determination wavelength was Hydroxysafflor yellow A at 400nm,Paeoniflorin at 235nm,Salvianolic acid B at 280 nm,Ferulic acid and Ligustilide at 324nm.Results:Five components were separated clearly and respectively in 24min.The relationship between the concentration and the peak areas of the five compounds was all linear.The RSDs of recovery,precision and reproducibility were all less than 2.0%.Conclusion: The method is rapid,simple,reliable and accurate,has been successfully used to the quantification of five compounds above mentioned in Naoxintong capsule.
