HPLC法测定野菊花提取物中3种二咖啡酰奎宁酸的含量

【摘要】 目的建立野菊花提取物中3种二咖啡酰奎宁酸的HPLC方法。方法 采用大连依利特HypersilODS2(4.6mm×250mm,5μm)色谱柱;以乙腈(A)-2%醋酸水溶液(B)为流动相,梯度洗脱(0~15min,10%A→15%A;15-25min,15%A→16%A;25-30min,16%A→60%A;30-40min,60%A→10%A);流速1.0mL·min-1;检测波长326nm;柱温35℃。结果 1,3-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸及3,4-二咖啡酰奎宁酸在0.0625-2.0000μg范围内线性关系良好(r>0.9990,n=6);平均回收率(n=6)分别为102.172%,101.120%,100.520%。结论 本方法操作简单,准确性和重现性良好,可以作为同时测定野菊花中3种二咖啡酰奎宁酸含量的方法。
【Abstract】 OBJECTIVE To establish a HPLC method for determination of three dicaffeoylquinic acids in extracts from Chrysanthemum indicum L.METHODS The separation was carried out on a Elite Hypersil ODS2 ( 4.6 mm × 250 mm,5 μm) colum eluted with mobile phases containing acetonitrile (A)-2% acetic acid solution (B) with gradient mode (0~15 min,10%A→15%A;15-25 min,15%A→16%A;25-30 min,16%A→60%A;30-40 min,60%A→10%A) at the flow rate of 1.0 mL·min-1.The column temperature was 35℃ and the detection wavelength was 326 nm.RESULTS Good linearity between the peak area and injected amount exists when the amount is within 0.0625-2.0000 μg (r > 0.9990,n = 6) for 1,3-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid and 3,4-dicaffeoylquinic acid.The average recoveries (n = 6) were 102.172%,101.120% and 100.520%,respectively.CONCLUSION The method is simple,accurate and repeatable and can be used for the determination of three dicaffeoylquinic acids simultaneously in Chrysanthemum indicum L.
