左旋多巴甲酯对剥夺性弱视猫外侧膝状体中一氧化氮合酶表达的影响

【摘要】 目的:观察左旋多巴甲酯对剥夺性弱视猫的治疗效果及其对外侧膝状体(LGN)神经元型一氧化氮合酶(nNOS)表达的影响。方法:健康2周龄幼猫30只随机分成6组:左旋多巴甲酯高、中、低剂量组,左旋多巴阳性组、模型组及正常对照组,每组5只。除了正常组,于4周时参照Hubel[1]经典实验方法对各组幼猫进行麻醉后的左眼上下睑缝合建立剥夺性弱视模型,在12周后打开缝合眼并开始给药,ig给与左旋多巴甲酯20、40、80mg.kg-1,阳性对照组给与左旋多巴40mg.kg-1,正常组与模型组为等剂量生理盐水,每天一次持续30d。采用免疫组化技术检测外侧膝状体中nNOS免疫阳性细胞表达差异。结果:与模型对照组比较,治疗各组LGN中相对应区域的nNOS免疫阳性细胞则有显著的增加(P<0.01和P<0.05)。结论:左旋多巴甲酯促进剥夺性弱视猫的外侧膝状体通路的功能恢复,增加内源性nNOS的表达来恢复弱视的视觉功能。

【Abstract】 Objective:To observe the effects of L-dopa methyl ester(LDME) on the expression of apoptosis and nNOS protein in neurons of the lateral geniculate nucleus of cats with monocular deprivation,and explore the therapeutic effect of L-dopa methyl ester on amblyopia and its action mechanism.Methods:30 normal cats were randomly divided into 6 groups,5 cats in each group:low dose(LDME L),medium dose(LDME M),and high dose of L-dopa methyl ester(LDME H),positive control group(PC),normal control group(NC) and model control group(MC).Referring to the classic experiments of Hubel [1],the kittens in each group were anesthetized and visually deprived by suturing the lids of the left eyes to perform the monocular deprivation amblyopia(the normal control group was excluded),then eye closure was open and treat with drug after 12 weeks.L-dopa methyl ester(20 mg.kg-1,40 mg.kg-1 g,80 mg.kg-1),L-dopa(40 mg.kg-1) and normal saline were given respectively to the corresponding animals for 30 days.The HE staining were checked the cats’ LGN features.The immunohistochemistry technique was used to detect the expression of nNOS.Results:Compared with the model control group,the number of nNOS immunoreactive cells of the administration of groups in the LGN layers were significantly increased(P<0.01 and P<0.05).Conclusion:L-dopa methyl ester can effectively improve conduction and visual function in model of amblyopic cat,the mechanism may be related to the increased contents of L-dopa methyl ester into the LGN layers and regulation of the endogenous nNOS expression.
