【摘要】 目的比较家兔静注与口服左氧氟沙星后血浆和房水药动学。方法选取54只家兔,分别静注或口服左氧氟沙星24mg﹒kg—1,高效液相色谱法测定血浆和房水中药物浓度,3p97软件计算药动学参数。结果静注与口服血浆中t1/2β/t1/2ke分别为1.42±0.29h和2.99±0.20(hP<0.01);AUC(0→t)分别为42.56±3.71μg·h·mL-1和33.48±2.98μg·h·mL-1(P<0.01)。房水中左氧氟沙星t1/2β/t1/2ke分别为2.53±0.65h和2.68±0.70h;tmax分别为0.50±0.00h和0.67±0.24h(P<0.05);Cmax分别为4.93±0.83μg·mL-1和1.59±0.38μg·mL-1(P<0.01)。结论左氧氟沙星口服给药可在血浆和房水达较高药物浓度,但静注更为明显,二者有明显差别。更多还原

【Abstract】 OBJECTIVE:To compare pharmacokinetics of levofloxacin in plasma and aqueous humor of rabbits after oral or intravenous administration.METHOTS:54 rabbits were randomised into two grops,oral or intravenous administration 24mg·kg -1 levofloxacin,respectively.The concentrations of plasma and aqueous humor were measured by HPLC.Pharmacokinetics parameter were calculated with 3p97.RESULTS:Results of intravenous administration or oral showed that the half-life (t 1/2 β/ke) of levofloxacin in plasma were 1.42±0.29h and 2.99±0.20h(P<0.01);the AUC (0→t) were 42.56±3.71 μg·h·mL -1 and 33.48±2.98μg·h·mL -1 (P<0.01),respectively.The t 1/2 β of levofloxacin in aqueous humor were 2.53±0.65h and2.68±0.70h;the t max were0.50±0.00 hand 0.67±0.24h(P<0.05);the peak concentrations (Cmax) were 4.93±0.83 μg·mL -1 and 1.59±0.38 μg·mL -1 (P<0.01),respectively.CONCLUSION:Levofloxacin showed higher level plasma and aqueous humor after oral administration,but there were significant different in t max or C max or AUC (0→t) between oral and intravenous administration.更多还原
