【摘要】 目的:建立测定人血浆中替米沙坦的方法。方法:以地西泮(DZP)为内标,无水乙醚为萃取溶剂,采用HYPERSIL C18柱(5μm,200mm×5.0mm)为分析柱,甲醇-磷酸盐缓冲液(0.01mol·L-1NH4H2PO4,用H3PO4调pH至3.0左右)(V/V)=70﹕30为流动相,流速1.0mL·min-1;检测波长为228nm,柱温为室温。结果:替米沙坦血药浓度线性范围为0.1155～7.3920μg·mL-1(r=0.9998),最低定量浓度为0.1155μg·mL-1;高、中、低3种浓度TMST的方法回收率为87.12%～108.22%;日内、日间RSD分别为2.54～8.09%和4.90～9.09%。结论:所建方法的灵敏度、准确度及回收率均能达到人体血浆药物浓度测定的要求,也可用于临床人体药动学的研究。更多还原

【Abstract】 Objective:To determine the telmisartan in human plasma.Methods:A liquid-liquid extraction method with absolute ether was used and diazepam was chosen as internal standard.The analytes were separated on a HYPERSIL C 18 column (200mm× 5.0mm,5μm) column at room temperature and detected at 228 nm.The mobile phase consisted of methanol–phosphate buffer (0.01moL·L -1 NH4H2PO4,adjusted pH to about 3.0 by H 3 PO 4;70:30) with a flow rate of 1.0 mL·min-1.Results:A good linearity was obtained from 0.1155 to 7.3920 μg·mL -1 in plasma with a correlation coefficient of 0.9998.The lowest concentration detected was 0.1155 μg·mL -1.The recovery of method were from 87.12% to108.22%.The within-day and between-day RSD were 2.54～8.09% and 4.90～9.09%,respectively.Conclusion:The method is sensitive,accurate and a good recovery,and can be used for determining clinical drug concentration and pharmacokinetic study.更多还原
