三七总皂苷与比索洛尔对急性心梗后左室重构大鼠心功能作用的比较

【摘要】 目的比较三七总皂苷(PNS)与比索洛尔对急性心肌梗塞(AMI)后左室重构(LVRM)大鼠心功能作用。方法除假手术组外采用结扎大鼠左冠状动脉前降支的方法建立AMI模型,术后24h后随机分为对照组和实验组,连续4w分别灌胃给予生理盐水2mg·kg-1、比索洛尔25mg·kg-1和PNS40、80、120mg·kg-1,观察PNS和比索洛尔对病鼠室间隔舒张末期厚度(IVSd)、左室舒张末期内径(宽度)(LVIDd)、室间隔收缩末期厚度(IVSs)、左室收缩末期内径(宽度)(LVIDs)、左室后壁舒张末期厚度(LVPWd)、左室后壁收缩末期厚度(LVPWs)、左室射血分数(EF)、左室收缩百分率(FS)、二尖瓣口舒张早期血流速度(MV)、心率(HR)等反映心功能变化指标的影响。结果 PNS高、中剂量组与比索洛尔组的MV、EF均显著上升(P<0.01),比索洛尔组及中剂量PNS组的FS显著上升、LVIDd和LVIDs均显著减小(P<0.01~0.05)。结论 PNS及比索洛尔均具有改善AMI后LVRM病鼠心功能的作用,可用于治疗心梗后的左室重构。

【Abstract】 Objective:To compare the effect of improving the cardiac function in rats with acute myocardial infarction-left ventricular remodeling(AMI-LVRM) between Panax Notoginsenoside (PNS) and Bisoprolol.Methods:Rat models of AMI were produced by ligation of left anterior descending coronary artery except the sham-operation group,building rat model of AMI,after 24 hours of postoperative in rats.The rats were randomly divided into control and experimental groups,thenrespectively gavaged NS,bisoprolol group and the low、middle ane high dosage of PNS for four consecutive weeks.The cardiac function index such as Ventricular septalend-diastolic thickness (IVS d),Left ventricular end-diastolic dimension(LVID d),End-systolic ventric -ular septal thickness (IVSs),Left ventricular end-systolic diameter(LVIDs),End-diastolic posterior wall thickness of left ventricular(LVPW d),End- systolic posterior wall thickness of left ventricular (LVPWs),Ejection fraction (EF),The percentage of left ventricular systolic(FS),Mitral early diastolic flow velocity mouth(MV),Heart rate(HR) in the rats were observed.Result:Compared with the model group,EF and MV(P<0.01) can be significantly improved by high dose of PNS group,middle dose of PNS group and Bisoprolol group;FS increased remarkablely(P<0.01~0.05),while LVID d and LVID s decreased significicantly(P<0.01~0.05)in the middle dose of PNS group and Bisoprolol group.Conclusion:PNS and Bisoprolol can improve the cardiac function in post AMI LVRM,and they can be used to treat the left ventricular remodeling after acute myocardial infarction.
