多糖结构修饰对抗肿瘤活性影响的研究进展

【摘要】 多糖是一种重要的生命物质,生物活性丰富,而且无毒副作用,有利于研发新药。探讨多糖的结构修饰与抗肿瘤活性的关系。通过对多糖分子采用增加基团和分子量降解的方法来进行结构修饰(亲水性基团的增加、分子量的降低)。增加基团主要是对主链或支链进行化学修饰,如烷基化、硫酸化、羧甲基化、磷酸化、棕榈酰化、磺酰化等化学修饰方法;分子量降低主要通过生物方法、物理方法、化学方法对多糖进行修饰。由于多糖的结构与多糖的抗肿瘤活性的联系紧密,如多糖的理化性质、黏度、溶解度、糖链、官能团、空间结构等。利用多糖的结构修饰方法,适当的改变多糖的结构这一单一因素,其抗肿瘤活性发生改变。多糖的糖链、官能团、空间结构、理化性质、溶解度、黏度等发生变化会对多糖的抗肿瘤活性产生重要影响。对多糖进行适当的结构修饰后,多糖的抗肿瘤活性普遍提高。对其进行系统地研究将会推进多糖药物的发展。但是多糖的结构与药理作用机制之间的关系有待进一步探讨。

【Abstract】 Polysaccharide is an important life matter,rich in biological activity and toxic side effects.It is conducive to development of new drugs.To explore the relationship between structural modification of polysaccharides and antitumor activities.Increase of groups and degradation of molecular weight were used to modify polysaccharides molecular structure. Hydrophilic groups increases,or the molecular weight reduces.Jncrease in the main group is the main chain or branched-chain chemical modification,such as alkylation,sulfation, carboxymethylation,phosphorylation,palmitoylation,and sulfonylation;lower molecular weight, mainly through biological methods,physical methods,chemical modification of polysaccharides. Because the structure of polysaccharides and polysaccharide anti-tumor activity are closely connected,.such as physical and chemical properties of polysaccharides,viscosity,solubility,sugar chain,functional groups,spatial structure._Polysaccharide structures using modified,an appropriate change of the single factor structure of polysaccharides changes anti-tumor activity. Polysaccharide chains,functional groups,spatial structure,physical and chemical properties, solubility and viscosity played important roles in anti-tumor activity of polysaccharide.After appropriate structure modification,anti-tumor activity of polysaccharide was increased significantly.Development of polysaccharides would be enhanced by systematical research. However,the role of polysaccharide structure and pharmacological relationship between the mechanisms need to be further explored.
