【摘要】 肉从蓉属植物是我国植物资源中的重要类群。肉从蓉具有补肾、益精、润肠、抗衰老等功效,素有"沙漠人参"之称。然而,近几十年来对肉从蓉的过度采挖,致使肉从蓉中药资源濒临枯竭,已被列为国家二级保护植物。目前对肉苁蓉的研究都集中在药用成分的研究,为了更好的揭示肉苁蓉的遗传多样性,本文利用ISSR方法分析了13个肉苁蓉居群的遗传多样性。分析表明:1.7个ISS¹⁸R引物共扩增出63个位点,其中多态性位点62个,多态位点百分率为98.44%。表明肉苁蓉具有较高的遗传多样性。2.利用Shannon指数与Nei指数估算13个居群肉苁蓉的遗传变异,肉苁蓉居群Shannon指数的变化范围在0.0866-0.3192之间,Nei指数的变化范围在0.0625-0.2302之间,所有居群根据Nei指数排列的顺序与根据Shannon指数排列的顺序基本一致。3.根据Nei法计算出肉苁蓉13个居群分化指数Gst0.6239,基因流系数Nm,为0.3015。表明总的遗传变异中有62.39%的变异存在于居群间,居群间的遗传变异占相当大的比重;基因流Nm=0.3015<1,说明居群存在明显的分化。更多还原

【Abstract】 Herba Cistanche is an important group in China plants resources.Cistanche decerticola is a common traditional Chinese medicine that has been used to reinforce the vital function of kidney, induce laxation and impotence,female infertility with a reputable name as "Desert Ginseng".However,because of over-excavation,this resource will die away in recent decades and has been listed in Second Class State Protection of Plants in China. At present,most study of C.decerticola focus on medicinal ingredients.In this article we apply ISSR to study 13 populations to reveal genetic diversity of C.decerticola. The ISSR molecular marker results were as follows: 1.ISSR analysis of genetic relationship for 13 populations of C.decerticola carried out with 7 ISSR primers.63 loci which contains 62 polymorphic loci were found and the percentage of polymorphic loci was 98.44%,indicating a high genetic diversity of Cistanche. 2.Genetic variation of F.Manshurica was counted in this paper with Shannon information index and Nei’s gene diversity index.The range of Nei’s index was from 0.0625 to 0.2302,and that of Shannon’ index was from 0.0866 to 0.3192,The variations of 13 populatons indicated by the two indexes were identical. 3.The coefficient of gene differentiation(Gst) is 0.6239.There are 62.39%of total variation among populations.The estimate of gene flow from Gst(Nm) is 0.3015,indicating obvious differentiation existed among 13 populations.更多还原
