黄精低聚糖的分离纯化及理化性质研究

【摘要】 目的:研究黄精低聚糖的分离纯化技术,并进行理化性质的分析。方法:采用80%乙醇回流提取黄精中小分子糖,然后通过D101大孔吸附树脂柱、活性炭柱层析、SephadexLH-20凝胶柱层析分离纯化,得灰白色粉末状黄精低聚糖(RPO-1),对其进行理化性质鉴别和总糖含量测定。结果:RPO-1为糖类化合物,样品中不含酚类物质、淀粉、糖醛酸、大分子蛋白质、多肽和核酸,用苯酚-硫酸法测定其总糖含量为91.29%。结论:黄精低聚糖的分离纯化方法有效、实用,为进一步开发应用提供了技术方法和理论依据。
【Abstract】 AIM:To study the isolation, purification, physical and chemical characters of oligosaccharide from Rhizoma polygonati. METHODS:The low molecular weight saccharide was reflux extracted with 80% ethanol, and then the RPO-1 sample was isolated and purified by D101 macroporous adsorption resin column, activated carbon column chromatography and Sephadex LH-20 gel column chromatography, and further studied on the physical and chemical properties identification and total sugar content determination. RESULTS:RPO-1 sample is carbohydrate and not contain phenols, starch, uronic acid, protein, polypeptide and nucleic acids. We measured the contents of total sugar was 91.29% by phenol-sulfuric acid method. CONCLUSION:Methods of isolation, purification and analysis were effective and practical for the preparation of Rhizoma polygonati oligosaccharide.
