BN大鼠与Wistar大鼠Ⅰ型超敏反应敏感性的比较

【摘要】 目的比较BN大鼠和Wistar大鼠在Ⅰ型超敏反应中的敏感性,建立一种灵敏可靠的Ⅰ型超敏反应检测体系。方法 Wistar大鼠和BN大鼠分别隔天皮下注射不同剂量(10、20、40μg·kg-1)的卵清蛋白,共5次,对照组用生理盐水代替。第21天取血清用ELISA法测定血清总IgE水平,并应用被动皮肤过敏实验检测其特异性IgE表达;第22天检测激发后大鼠血压、血清中组胺和类胰蛋白酶浓度的变化。结果与对照组相比,BN大鼠在10、20和40μg·kg-1剂量下血清总IgE和特异性IgE显著增加,血压下降,血清组胺和类胰蛋白酶浓度增加;而Wistar大鼠仅在40μg·kg-1剂量出现上述症状。结论与Wistar大鼠相比,BN大鼠用于Ⅰ型超敏反应的检测更为灵敏;血清总IgE、特异性IgE、血压、组胺和类胰蛋白酶等可作为Ⅰ型超敏反应重要的检测指标。

【Abstract】 AIM To compare the sensitivity in type Ⅰ hypersensitivity between Wistar and BN rats and to establish a sensitive and reliable determination system for type Ⅰ hypersensitivity. METHODS Wistar and BN rats were given different doses of ovalbumin (10, 20, 40 μg·kg-1) subcutaneously every other day for a total of five injections and the control group was injected normal saline. The total IgE levels in serum were determined by ELISA and the specific IgE levels in serum were determined by passive cutaneous anaphylacxis on the 21th day. The blood pressure,histamin levels and tryptase levels in serum were determined after challenge on the 22th day. RESULTS Total IgE, specific IgE, histamine and tryptase levels all significantly increased and blood pressure decreased in doses of 10, 20 and 40 μg/kg in BN rats compared with the control group, while in Wistar rats these symptoms only appeared in dose of 40 μg·kg-1. CONCLUSION BN rats are more sensitive than Wistar rats in type Ⅰ hypersensitivity. The total IgE, specific IgE, blood pressre, histamin levels and tryptase levels can be used important indicators in type Ⅰ hypersensitivity.
