反相高效液相法测定人体血浆中拉莫三嗪的浓度

【摘要】 目的:建立人血浆中抗癫痫药拉莫三嗪的RP-HPLC测定方法。方法:以萘普生为内标,血浆样品以直接蛋白沉淀法处理。采用XTerra?RP18柱(5μm,4.6mm×250mm),流动相为甲醇-0.01mol·L-1磷酸二氢钾缓冲液(pH6.0)(66:34),流速为1.0mL·min-1,检测波长为280nm。结果:拉莫三嗪的血药浓度在0.5~32.0mg·L-1范围内线性良好,回归方程为Y=0.1389X-0.0232(r=0.9999),最低检测浓度为0.2mg·L-1,提取回收率为68.20%~82.35%,方法回收率为99.03%~103.21%,日内RSD、日间RSD均<4.00%。结论:该方法操作简便,灵敏度高,重复性好,能快速准确测定人血浆拉莫三嗪浓度。

【Abstract】 OBJRCTIVE:To develop method for determination of lamotrigine in human plasma by a RP-HPLC. METHODS:The analytes were extracted by direct protein precipitation, using naproxen as internal standard. XTerra?RP18 column (250mm×4.6mm, 5μm) was used and the mobile phase was composed of methanol-0.01mol ·L-1 potassium dihydrogen phosphate buffer(pH 6.0)(66:34), The flow rate was 1.0mL·min-1, the detection wavelength of UV was 280nm. RESULTS:A good linear calibration curves was obtained at the range of 0.5~32.0 mg·L-1. The regression equation was Y = 0.1389X - 0.0232(r = 0.9999). The minimum detection was 0.2 mg·L-1 and the extraction recovery was 68.20%-82.35% and the method recovery was 99.03%~103.21%. The RSD of intra-day and inter-day were all blow 4.00%. CONCLUSION:The method is simple, sensitive, accurate and reproducible for concentration determination of lamotrigine in human plasma.
