三七总皂苷对心梗后心室重构大鼠TNF-α与MMP-2的表达影响及其作用机制

【摘要】 目的研究三七总皂苷(PNS)对急性心肌梗塞(AMI)后左室重构(LVRM)大鼠血清肿瘤坏死因子-α(TNF-α)与基质金属蛋白酶-2(MMP-2)的表达影响并探讨其作用机制。方法除假手术组外采用结扎大鼠左冠状动脉前降支的方法建立AMI模型,术后24h后随机分为对照组和实验组,连续4周分别灌胃给予生理盐水、福辛普利和PNS低、中、高剂量,观察PNS对病鼠左室舒张末期内径(宽度)(LVIDd)、左室收缩末期内径(宽度)(LVIDs)、左室射血分数(EF)、左室收缩百分率(FS)、二尖瓣口舒张早期血流速度(MV)、心率(HR)等反映心功能变化指标及TNF-α与MMP-2表达的影响。结果 PNS中、高剂量组的EF、FS及MV指标均显著提高(P<0.01~0.05);PNS低、中、高剂量组均能明显抑制TNF-α与MMP-2的表达(P<0.05)。结论 PNS能够抑制TNF-α与MMP-2的分泌与释放,通过提高左室收缩和舒张功能,降低外周阻力,减轻AMI后LVRM大鼠的心肌缺血再灌注损伤,对病鼠AMI后LVRM心功能有保护和改善作用。

【Abstract】 OBJECTIVE Research on effects and mechanism of Panax Notoginsenoside(PNS) on TNF-α and MMP-2 in rats with post-myocardial infarction ventricular remodeling. METHODS Models of AMI were produced by ligation of left anterior descending coronary artery, building rat model of AMI, after 24 hours of postoperative in rats.The rats were randomly divided into control and experimental groups,then respectively gavaged NS 、fosinopril and the low、middle and high dosage of PNS for four consecutive weeks. To observe effects of PNS on the cardiac function index such as Left ventricular end-diastolic dimension(LVIDd), Left ventricular end-systolic diameter (LVIDs), Ejection fraction(EF),The percentage of left ventricular systolic(FS), Mitral early diastolic flow velocity mouth(MV),Heart rate(HR) and TNF-α and MMP-2 in rats with post-myocardial infarction ventricular remodeling. RESULT Compared with the NS group:EF,FS and MV(P <0.01~0.05) can be significantly improved by PNS in middle and high dosage group.TNF-α and MMP-2 (P <0.01) can be significantly decreased by PNS in low、middle and high dosage groups. CONCLUSION PNS can inhibit or reduce the expression of TNF-α and MMP-2, which enhanced left ventricular systolic and diastolic function, decreased peripheral resistance,and improve the cardiac function in post AMI LVRM.
