卷柏总黄酮通过调节糖尿病大鼠胰腺内分泌长效降糖的实验研究

【摘要】 目的研究卷柏总黄酮对二型糖尿病大鼠一般情况、血糖水平、抗氧化水平及对胰岛素和胰高血糖素调节的影响,探讨卷柏总黄酮治疗糖尿病的特点。方法雄性Wistar大鼠以高糖高脂饲养以诱发胰岛素抵抗,4周后腹腔注射35mg/kg链脲佐菌素(STZ)以建立二型糖尿病模型。连续给药8周,检测体重、摄食量、饮水量、空腹血糖(FBG)、糖化血红蛋白(HbA1c)、口服葡萄糖耐量(OGTT)、胰岛素、C-肽、胰高血糖素。结果卷柏总黄酮100mg/kg、200mg/kg、400mg/kg均能不同程度地降低FBG、HbA1c,改善OGTT,调节胰岛素和胰高血糖素分泌,增高SOD,降低MDA,提高机体抗氧化能力,并使体重维持恒定。结论卷柏总黄酮通过调节胰腺内分泌,长期控制血糖稳定。
【Abstract】 OBJECTIVE To investigate the effects of total flavones in Selaginella tamariscina (TFST) on general index, blood glucose levels, antioxidant abilities,regulation of insulin and glucagon, to explore the distinctive features of TFST in treatment of diabetes. METHODS Male Wistar rats were fed with high fat/sugar diet to induce insulin resistant. After 4 weeks of dietary manipulation, the rats were intraperitoneally injected with streptozotocin (STZ) 35mg/kg to induce type Ⅱdiabetic model. The duration of administration was 4 weeks, and then the body weight, food intake, water intake were recorded, the levels of fast blood glucose (FBG) and glycosylated hemoglobulin A1C (HbA1c) were measured , oral glucose tolerance test (OGTT) was detected and the serum levels of insulin, C-peptide and glucagon were observed. RESULTS TFST (100mg/kg、200 mg/kg、400mg/kg) could decrease the serum levels of FBG and HbA1c in different degrees, improve OGTT, regulate the insulin and glucagon, reduce the content of MDA, increase the activity of SOD and help maintain a stable body weight. CONCLUSION TFST can promote long-term stabilization of blood glucose levels by regulating the endogenous secretions of pancreas.
