人参皂苷Rh3对人结肠癌细胞增殖与凋亡的影响

【摘要】 目的:探讨人参皂苷Rh3对人结肠癌细胞SW480增殖与凋亡的影响。方法:分别用15、30、60、120和240μg/ml的人参皂苷Rh3作用于人结肠癌细胞SW48024、48及72h,MTT比色法检测细胞增殖;用15μg/ml的人参皂苷Rh3作用24及72h,TUNEL法检测细胞凋亡;用15和20μg/ml的人参皂苷Rh3作用24及72h,丫啶橙/溴化乙啶双染法计算细胞凋亡率和坏死率。结果:人参皂苷Rh3可抑制SW480细胞增殖,其作用随剂量增加和作用时间延长而增强,30μg/ml人参皂苷Rh3作用72h抑制率超过80%;15μg/ml人参皂苷Rh3作用24h细胞凋亡率为25.5%,20μg/ml人参皂苷Rh3作用24h细胞凋亡率为31%,未见细胞坏死,15μg/ml人参皂苷Rh3作用72h细胞凋亡率为55%、坏死率为7%,20μg/ml人参皂苷Rh3作用72h细胞凋亡率为67.5%、坏死率为7%。结论:人参皂苷Rh3能抑制人结肠癌细胞SW480增殖,诱导其凋亡,作用呈剂量依赖性和时间依赖性。
【Abstract】 OBJECTIVE To investigated the effect of ginsenoside Rh3 on proliferation and apoptosis of human colon carcinoma cells SW480. METHODS Human colon carcinoma cells SW480 were cultured in vitro and treated with 15, 20, 30, 60, 120 and 240 μg/ml of ginsenoside Rh3 for 24, 48 and 72 h, respectively. The proliferation of SW480 cells was determined with MTT colorimetric method. TUNEL and acridine orange/ethidium bromide double staining were used to detect cell apoptosis and calculate apoptosis ratio and necrosis ratio. RESULTS 15 μg/ml of ginsenoside Rh3 treating 48 h was effective to inhibit SW480 cell proliferating, and the effect was enhanced with dose increasing and time prolonging. The inhibition ratio was above 80% when the cells were treated with 30 μg/ml of ginsenoside Rh3 for 72 h. The result of TUNEL showed that apoptotic cells was corrugativus and brow-stained in cytoplasm. The number of apoptotic cell treated for 72 h was higher than that treated for 24 h. The apoptosis ratio was 25.5% of the cells treated with 15 μg/ml of ginsenoside Rh3 for 24 h and 31% with 20 μg/ml of ginsenoside Rh3. The apoptosis ratio was 55%, and the necrosis ratio was 7% of the cells treated with 15 μg/ml of ginsenoside Rh3 for 72 h. The apoptosis ratio was 67.5%, and the necrosis ratio was 7% of the cells treated with 20 μg/ml of ginsenoside Rh3 for 72 h. CONCLUSIONS Ginsenoside Rh3 could inhibit human colon carcinoma cell SW480 proliferating and induce apoptosis in a dose-dependent and time-dependent pattern.
