灯盏花乙素苷元对实验性脑缺血的保护作用

【摘要】 目的:研究灯盏花乙素苷元对大鼠实验性脑缺血的保护作用。方法:Wistar雄性大鼠分别以100,50,25mg·kg-1的灯盏花乙素苷元连续灌胃给药7天,采用电凝法阻断大脑中动脉(MCAO)制备大鼠局部脑缺血模型,缺血24h后对大鼠神经功能进行评分,并测定脑梗死面积;采用结扎双侧颈总动脉致全脑缺血模型,缺血6h后观察脑组织病理改变,缺血48h后取脑组织测定脑含水量和脑组织中Na+、K+含量;电刺激大鼠颈动脉复制动脉血栓模型,记录血栓形成时间;结扎大鼠下腔静脉复制静脉血栓模型,记录血栓湿重和干重;采用二磷酸腺苷(ADP)诱导血小板聚集,测定最大聚集率;大鼠皮下注射肾上腺素复制血瘀模型,测定红细胞沉降率、全血表观粘度和血浆粘度。同时设灯盏花乙素(100mg·kg-1)阳性对照。结果:灯盏花乙素苷元能够剂量依赖性地改善MCAO局部脑缺血大鼠的神经功能障碍,缩小脑梗死面积;降低全脑缺血大鼠的脑含水量,抑制缺血脑组织的Na+潴留及K+流失现象,改善光镜下脑组织缺血性损伤;并能够明显延缓大鼠动脉血栓形成时间,抑制静脉血栓形成,抑制ADP诱导血小板聚集,降低血瘀模型大鼠的红细胞沉降率和血粘度。结论:提示灯盏花乙素苷元对大鼠实验性脑缺血模型具有保护作用,疗效优于灯盏花乙素。
【Abstract】 Objective:To investigate the protective effect of scutellarein on rat cerebral ischemia. Method:After oral administration of scutellarein 100, 50 or 25mg?kg-1 for 7 days, focal cerebral ischemia was induced in male wistar rats by middle cerebral artery occlusion with the method of electric coagulation, and 24 hours later, the neurological deficit score and the cerebral infarct volume were assessed. Besides, in rats undergone same medication schema, global cerebral ischemia was induced by ligation of bilateral carotid and the pathological changes were assessed meanwhile the water content and Na+, K+ concentration in brain were also measured. Furthermore, thrombosis time of carotid artery under electrical stimulation, thrombus weight under inferior vena cava ligation and platelet aggregation rate induced by ADP were observed. The blood stasis model in rats was established by hypodermic injection of epinephrine, and erythrocyte sedimentation rate, whole blood and plasma viscosities were measured. In all experiments, 100 mg·kg-1 scutellarin was tested as positive control. Results:Scutellarein could dose dependently ameliorate the neurological behavior and restrict the infarction development in MCAO rats. Also, it could reverse the brain edema, the ionic disorder and the pathological changes in global cerebral ischemia. In addition, scutellarein could clearly depress arterial or venous thrombosis, restrict ADP-induced platelet aggregation rate,reduce ESR, whole blood and plasma viscosities in blood stasis rats. Conclusion:Scutellarein has protective effect, better than scutellarin, in rat cerebral ischemia.
