SA14-14-2株乙型脑炎减毒活疫苗细胞免疫持久性研究

【摘要】 目的研究SA14-14-2株乙脑减毒活疫苗在小鼠体内的细胞免疫持久性。方法以SA14-14-2株乙脑减毒活疫苗0.1ml皮下免疫BALB/c小鼠,分别于第14天、1个月、3个月、6个月和一年处死,取脾脏,ELISPOT法检测脾淋巴细胞经乙脑活疫苗抗原刺激后分泌IFN-γ和IL-2的水平,同时分离血清,混合后检测中和抗体水平。另一组相同免疫的小鼠于免后各时间点以P3毒株进行腹腔攻击,观察疫苗保护效果。结果乙脑减毒活疫苗免后14天细胞免疫水平达到高峰,持续1~3个月后开始略有下降,免后3个月至1年间的脾细胞IFN-γSFC和IL-2SFC数量仍维持在较高水平。小鼠中和抗体水平在3个月内由1:20增加到1:80,一年内仍维持在1:40。1年内乙脑减毒活疫苗免疫的小鼠均能很好地抵御乙脑强毒株的腹腔攻击。结论 SA14-14-2株乙脑减毒活疫苗能同时诱导细胞免疫反应和体液免疫反应,细胞免疫至少能维持一年,活疫苗产生的保护效果也至少能持久一年。
【Abstract】 Objective To study the cellular immune persistence of live attenuated SA14-14-2 Japanese encephalitis vaccine in mice. Methods Live attenuated SA14-14-2 Japanese encephalitis vaccine was subcutaneously injected to BALB/c mice one time. Fourteen days, one month, three months, six months and one year after the immunization, mice were sacrificed, and the spleens were removed aseptically for detection of IFN-γ and IL-2 spot forming cell (SFC) by ELISPOT assay. Serum samples were collected to detect the neutralizing antibody. Another group of immunized mice were challenged by intraperitoneal injection of Japanese encephalitis virus P3 strain to evaluate the protective efficacy at the same time. Results The cellular immunity level reached the peak at 14 days after immunization with the live attenuated JE vaccine and then decreased after 1~3 months. The rates of IFN-γ SFC and IL-2 SFC in spleen cells were maintained at a certain level from 3 months to 1 year after immunization. Neutralizing antibody titers in mice increased from 1:20 to 1:80 by 3 months and kept at 1:40 in one year after immunization. The mice could be protected against the challenge of Japanese encephalitis virus by intraperitoneal route during one year after immunized with the live attenuated JE vaccine. Conclusion Live attenuated SA14-14-2 Japanese encephalitis vaccine could induce both humoral and cellular immune response. The cellular immune response could maintain at least one year. The protective effect induced by the live attenuated JE vaccine could last at least one year as well.
