菟丝子多糖超声提取工艺优化

【摘要】 目的:以多糖得率及还原力为指标,优化菟丝子多糖超声提取工艺。方法:先逐一考察脱脂所用乙醇浓度、超声提取时间、温度和料液比等因素对多糖得率及还原力的影响,再用正交试验综合分析上述各因素交互作用对多糖得率及还原力的影响。结果:菟丝子多糖得率及还原力受脱脂处理所用乙醇浓度、超声提取时的料液比和温度等影响较大,而受提取时间的影响较小。结论:菟丝子多糖超声提取的最佳工艺为:无水乙醇室温浸泡过夜后超声脱脂,40℃低温真空干燥,按料液比1∶15加蒸馏水,60℃水浴下超声提取80min。

【Abstract】 Objective:the yield and reducing power were employed as indexes to optimize the ultrasonic extraction condition of polysaccharide from Semen Cuscutae. Method:Firstly, factors of the ultrasonic extraction condition such as extraction time, temperature, solid-liquid ratio as well as concentration of ethanol used for defatting were investigated respectively. And then, orthogonal experiments were design and used to comprehensive analyse the interaction of the above 4 factors on the polysaccharides yield and reducing power. Result:the yield and reducing power of polysaccharides extracted from Semen Cuscutae were more affected by factors of the concentration of ethanol used for defatting, the solid-liquid ratio and the extraction temperature than by that of the extraction time. Conclusion:the optimal ultrasonic extraction condition is:After soaking in 100%(M/V) ethanol for overnight, the Semen Cuscutae was ultrasonic defatted and vacuum dried to constant weight, after adding 15ml distilled water per gram, the Semen Cuscutae was ultrasonic extracted for 80 min under 60 ℃.
