构建穿心莲内酯微乳给药系统
Preparation and quality evaluation of Andrographolide microemulsion

【摘要】 目的研制穿心莲内酯微乳给药系统,并评价其质量。方法绘制伪三元相图,根据三元相图优选微乳处方,研制微乳给药系统,考察载药微乳的外观、形态、粒径、电位及稳定性,用高效液相色谱法测定穿心莲内酯的含量、包封率和载药量。结果根据伪三元相图,确定穿心莲内酯微乳给药系统的处方如下:吐温80(Tween 80)为乳化剂,聚乙二醇600(PEG 600)为助乳化剂,油酸乙酯(EO)为油相;研制的穿心莲内酯微乳为淡黄色略带蓝色乳光的半透明液体,透射电镜下呈圆球形,分布均匀,用马尔文粒度检测仪测得微乳的平均粒径为34.41 nm,10000r·min-1离心20 min,微乳系统无分层现象,高效液相色谱法测定其包封率为90.81%,载药量为0.46 g·L-1。结论本研究构建的穿心莲内酯微乳给药系统,为热力学稳定的纳米体系,处方合理,制备简单,质量可靠。
【Abstract】 OBJECTIVE Andrographolide microemulsion was prepared and its quality was evaluated. METHOD:Andrographolide-loaded microemulsion was prepared by using the method of titration to prepare the pseudo-ternary phase diagram.The appearance,morphology,size distribution,Zeta potential and stability of Andrographolide microemulsion were determined.The concentration,encapsulated efficiency and the contents of Andrographolide were determined by HPLC.Andrographolide microemulsion was stable after high speed centrifugation and reserving at 4℃,room temperature and 60℃.RESULT:Tween 80,PEG 600,EO and distilled water were selected to prepare Andrographolide microemulsion.The Andrographolide microemulsion, with good stability,was a translucent diluted-yellow solution with visible sky-blue opalescence.Andrographolide microemulsion in electron microscopy consisted of small spherical drops with a mean diameter of 34.41nm.The encapsulated efficiency and the concentration of Andrographolide microemulsion were 90.81%and 0.46 g·L-1, respectively.CONCLUSION:The solubility of Andrographolide was significantly increased in Andrographolide microemulsion and the stability of microemulsion was satisfying,Andrographolide microemulsion is easy to prepare.
