【摘要】 目的注射用双黄连(冻干)(Shuanghuanglian,SHL)是由三味中药提取物组成的中药注射剂,广泛用于抗菌、抗病毒的治疗。本研究以大鼠为模型对其类过敏反应及机制进行了探讨。方法采集大鼠给药后的静脉平均动脉压及心电图,观察SHL对大鼠血压的影响,利用苯海拉明、化合物48/80、色甘酸钠对其降压机制进行研究并检测化合物48/80及SHL给药后血浆组胺水平。结果大鼠首次给予SHL和组胺均可导致血压下降,但对心电和心率无影响。苯海拉明可部分阻断SHL的降压作用。色甘酸钠可阻断化合物48/80的作用,但对SHL无阻断作用。静脉注射化合物48/80及SHL均可导致血浆组胺水平升高。结论SHL首次给药可导致大鼠血压明显下降,并可引起血浆组胺水平升高,提示其的作用机制可能与促进组胺释放或存在类组胺物质有关,为类过敏反应机制。更多还原

【Abstract】 OBJECTIVE Shuanghuanglian (SHL), a extract mixture isolated from three medicinal herbs, has been used for viral or bacterial infection as a Traditional Chinese Medicine Injection in China. Hypotensive responses in rats induced by SHL and the possible mechanism were examined. METHODS Mean arterial pressure (MAP) and electrocardiogram (ECG) was examined after intravenous injection of histamine and SHL. H1 receptor antagonist Diphenhydramine, histamine release promoting agent Compound 48/80 and histamine release inhibitor Cromolyn were also used by giving previously to investigate the potential mechanism. In addition, the histamine level in plasma was measured after administration of SHL and Compound 48/80. RESULTS Both SHL and histamine led to MAP reduction immediately but did not influence ECG after administration in a similar manner first contacted, which could be attenuated by Diphenhydramine partially. Pretreatment could block the reaction of Compound 48/80 but not that of SHL. And plasma histamine level was elevated by SHL. CONCLUSION SHL can provoke severe hypotension through histamine release at the first contacting, combined with direct effects of histamine similar materials on target tissue, and is highly suspected to own the potential of causing anaphylactoid reaction.更多还原
