应用正交设计法研究不同促渗剂对茶多酚乳膏经皮渗透的影响

【摘要】 目的:考察不同促渗剂对茶多酚乳膏的体外经皮渗透的影响,通过比较透过量和皮肤中药物含量,为该制剂的组方提供实验依据。方法:采用Franz扩散池,以小鼠离体皮肤作为渗透屏障,应用正交设计研究氮酮、尿素和丙二醇三种促渗剂对茶多酚乳膏经皮渗透的影响;以茶多酚中主要活性成分EGCG为检测指标,采用HPLC测定。结果:三种不同促渗剂虽可增加6%茶多酚乳膏的24h累积渗透量(顺序为:氮酮>丙二醇>尿素),但无统计学显著性;研究发现,氮酮能显著增强EGCG的皮内摄取(P<0.01),而尿素和丙二醇尽管有某种程度的增强作用,但无统计学意义。结论:2%氮酮明显增加6%茶多酚乳膏在皮肤的摄取,可作为促渗剂用于制备茶多酚乳膏。

【Abstract】 OBJECTIVE To study the effects of penetration enhancers on percutaneous penetration of tea polyphenol(TP)cream. METHODS Isolated mouse skin was used as permeation barrier. Using orthogonal design,the permeation–enhancing effect of azone,propylene glycol and urea was investigated in Frantz diffusion cell by HPLC determination of EGCG, a main active ingredient of TP, in receptor’s solution.RESULTS The 3 permeation enhancers appeared to increase the cumulative permeation quantity of EGCG through skin,ranked in order of azone>propylene glycol >urea,but there was no statistical significance(P>0.05).However,azone was found to have significant enhancing effects on intra-dermal uptake of EGCG ( P< 0.01 ), whereas propylene glycol and urea showed enhancing effect to a certain extant,but without statistical significance.CONCLUSION 2%Azone can obviously enhance the uptake of 6%TP cream within the whole skin and,therefore,is chosen as permeation enhancer to prepare tea polyphenol cream.
