大川芎方效应组分血清药物化学研究

【摘要】 目的:借鉴血清药物化学的方法,探讨大川芎方的药效物质基础,为有效、全面地控制大川芎方及其制剂的质量奠定基础。方法:通过HPLC指纹图谱和紫外光谱分析,比较空白血清、含药血清的成分异同,确定大川芎方效应组分的血中移行成分,并采用HPLC-DAD-MS分析技术推测移行成分的化学结构。结果:大川芎方效应组分给药后血中出现了4个移行成分,阿魏酸、洋川芎内酯I来自于川芎,而天麻素、巴利森苷来自于天麻。结论:通过研究大川芎方效应组分血中移行成分,可以快速、有效地确定川芎效应组分的物质基础,为进一步建立其完善的质量标准奠定基础。

【Abstract】 Objective To study the substance basis of the Dachuanxiong Fang active parts (DFAP) by serum pharmacochemistry, so that the quality of DFAP can be effectively and comprehensively controlled. METHODS Based on the HPLC fingerprints and UV spectrums, comparison was carried out among the peaks of the blank serum and drug-contained serum, through which the DFAP ingredients absorbed into blood post oral administration were confirmed followed by the structural identification using HPLC-DAD-MS. Results Four DFAP components detectable in rat bloods following the oral administration of DFAP were ferulic acid, senkyunolide I, gastrodin and parishin, among which the former two come from Chuanxiong and the latter two from Tianma. Conclusion The findings obtained from the study of absorbed DFAP constituents can facilitate the determination of bioactive substance basis of DFAP and the improvement of quality control of DFAP.
