溴隐停与罗格列酮和二甲双胍对PCOS大鼠胰岛素抵抗、血脂和氧化应激作用比较研究

【摘要】 目的:观察溴隐停、二甲双胍、罗格列酮分别对多囊卵巢综合征(PCOS)大鼠胰岛素抵抗、血脂和氧化应激作用。方法:采用十一酸睾酮加绒促性素(HCG)建立PCOS大鼠模型,连续口服给予溴隐停、二甲双胍、罗格列酮6周,观察大鼠体重、空腹血糖(FBG)、口服葡萄糖后2小时血糖(OGTT-2hBG)、胰岛素、血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、肿瘤坏死因子-α(TNF-α)、睾酮(T)、胰岛素样生长因子(IGF-1)、丙二醛(MDA)、一氧化氮(NO)等的含量,以及超氧化物歧化酶(SOD)和诱导型一氧化氮合酶(iNOS)、一氧化氮合酶(NOS)的活性,并计算胰岛素敏感指数(ISI)的变化。结果:PCOS大鼠体重,FBG,OGTT-2hBG,胰岛素水平,TG,TC,LDL-C,TNF-α,T,IGF-1,MDA,NO和iNOS均明显升高,而ISI,HDL-C和SOD显著降低,与空白对照组比较,差异有高度显著性(P<0.01);溴隐停、二甲双胍、罗格列酮均能不同程度地改善PCOS大鼠的上述病理改变(P<0.01~0.05)。结论:溴隐停、二甲双胍及罗格列酮均能不同程度地改善PCOS大鼠胰岛素抵抗(IR)、血糖和血脂代谢异常、高雄激素血症以及氧化应激状态,且各药作用强度间差异未见显著性。
【Abstract】 Objective:To investigate the effects of bromocriptine and rosiglitazone and metformin on insulin resistance metabolism of lipids and oxidative stress in rats with polycystic ovary syndrome(PCOS).Method:The model of PCOS rats induced by testosterone undecanoate and human chorionic gonadotropin(HCG),afterwards bromocriptine,rosiglitazone and metformin were taken by intragastric administration for 6 weeks.At the end of the experiment,the bodies weight of rats were measured,and the contents or activities of fasting blood-glucose(FBG),2-hours blood glucose after oral glucose tolerance test(OGTT-2 hBG),insulin,total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),Tumor necrosis factor(TNF-α),testosterone(T),resistin,insulin growth factor-1(IGF-1), malondialdehyde(MDA),nitric oxide(NO),inducible nitric oxide synthase(iNOS)and nitric oxide synthase(NOS)were determined,and the changes of insulin sensitivity index(ISI)were counted.Result:Compared with the normal group,the bodies weight of rats and concentrations or activities of FBG,OGTT-2hBG,insulin,TC,TG,LDL-C,TNF-α,T,resistin,IGF-1, MDA,NO and iNOS were increased obviously(P<0.01),but ISI,HDL-C and SOD were decreased significantly(P<0.01).All of drugs could improved the pathologic changes in PCOS rats(P<0.01or P<0.05),Conclusion:All of bromocriptine and rosiglitazone and metformin can improve the pathologic changes in PCOS rats on IR,blood glucose,blood lipid, hyperandrogenemia and oxidative stress state,but the differences among potency of drugs appear not significant.
