AP-1表达变化在黑米花青素防护大鼠视网膜光化学损伤中的作用

【摘要】 目的:从转录活化蛋白-1(Activator protein-1,AP-1)表达变化角度探讨黑米花青素(Anthocyanins-rich extract from black rice,BRACs)防护视网膜光化学损伤(retinal photochemical damage,RPD)的分子机制。方法:60只Sprague-Dawley大鼠饲以AIN-93基础饲料1周后,随机分为对照组(饲以基础饲料,同时3000±200lux强度的白色荧光持续光照24h,n=30)和BRACs组(饲以基础饲料的同时,给予100mg BRACs/kg体重灌胃15d,光照24h,n=30),RT-PCR法和免疫组化法检测视网膜内AP-1(c-fos/c-jun)mRNA和蛋白表达水平。结果:光照后c-fos和c-jun mRNA表达均增加,BRACs干预后c-fos和c-jun mRNA表达显著低于对照(p<0.05);光照后外核层和内核层出现大量的c-fos和c-jun阳性细胞,而BRACs干预后c-Fos和c-jun阳性细胞数明显减少。结论:BRACs可能通过抑制视网膜外核层和内核层c-Fos和c-Jun表达,减少AP-1复合物的形成,阻断感光细胞的凋亡诱导从而防护视网膜光化学损伤。
【Abstract】 Objective The present experiment is to investigate the molecular mechanism of dietary supplementation with anthocyanins-rich extract from black rice(BRACs) on retina photochemical damage based on the alteration of AP-1 expression in retina. Methods Sixty Sprague-Dawley rats fed with AIN-93 diet for 1 wk were randomly divided into control group(fed with AIN-93 diet and exposed to 3000 ± 200 lux fluorescent light for 24h,n=30) ,and BRACs group(intragastric administration with 100mg BRACs/kg body weight for 15d and exposed to light for 24h,n=30) . Thereafter,the alterations of c-Fos/c-Jun mRNA and protein expression were determined by RT-PCR and immunohisto-chemistry,respectively. Results The mRNA of c-fos and c-jun were increased after light exposure,and lower in BRACs group than that in control(p<0.05);After light exposure,there were considerable c-Fos/c-Jun positove cells. But the supplementation with BRACs decreased the number of c-Fos/c-Jun positove cells. Conclusion BRACs down-regulate the level of expression of c-Fos/c-Jun protein and mRNA in light-injured retina and may furthermore decrease the formation of AP-1 complex,which suppress the photoreceptor cells apoptosis and protect retina from photochemical damage.
