高脂饮食大鼠血清apelin及脂肪组织apelin mRNA表达与胰岛素抵抗关系的研究

【摘要】 目的:研究高脂饮食诱导的胰岛素抵抗大鼠血清apelin水平及其脂肪组织apelinmRNA表达。探讨apelin与体脂、糖、脂代谢、高胰岛素血症等的相关性。方法:清洁级健康雄性Wistar大鼠30只,体重290-300g。适应性喂养一周,随机分为正常对照组15只,高脂饮食组15只。正常对照组给予基础饲料(热量组成:碳水化合物占65.5%,脂肪占10.3%,蛋白质占24.2%),总热量为348 kcal/100g;高脂饮食组喂以高脂饲料(热量组成为脂肪59.8%,碳水化合物20.1%,蛋白质20.1%),总热量为501 kcal/100g。2组大鼠给予等量饲料。4周后2组大鼠禁食10小时过夜,内眦取血,分离血清,并行高胰岛素-正葡萄糖钳夹实验,测定大鼠葡萄糖输注率。确认造模成功后,测定2组大鼠血清apelin-36、空腹血胰岛素(Fins)、空腹血糖(FPG)、甘油三酯(TG)、总胆固醇(TC),游离脂肪酸(FFA)等指标。2组大鼠均放血处死,准确称量肾周和附睾脂肪重量,并分离肾周或附睾脂肪,采用半定量RT-PCR技术测定脂肪组织apelin mRNA表达。所得数据采用SPSS13.0统计软件处理,计量资料数据以±s表示。检验两组变量正态分布情况,组间比较用t检验,多因素分析采用Pearson相关及多元逐步回归分析。结果:4周时高脂组葡萄糖输注率低于正常对照组(p<0.01);高脂组apelin-36、FPG、Fins、腹内脂肪、TG、TC、FFA高于正常对照组(p<0.01,均有统计学差异),两组体重比较(p>0.05)无统计学差异;相关分析显示:血清apelin与Fins、FPG(r=0.497,r=0.481,均P<0.01)呈正相关,与TG、TC、腹内脂肪含量、体重、FFA呈正相关(r=0.355,r=0.324,r=0.339,r=0.220,r=0.191,p<0.05);在胰岛素抵抗大鼠中,以apelin为因变量,以Fins、FPG、体重、腹内脂肪、TG、TC、FFA为自变量做多元逐步回归分析,表明血浆胰岛素水平是apelin独立相关因素;脂肪组织RT-PCR显示大鼠脂肪组织apelin在高脂组脂肪组织表达相对含量高于正常对照组,两组比较P<0.01。结论:血清apelin水平及脂肪组织apelin mRNA表达在胰岛素抵抗大鼠中增高,血清apelin与高胰岛素血症、体脂、糖、脂代谢相关,血浆胰岛素水平是其独立相关因素,推测apelin可能参与了胰岛素抵抗的发生。

【Abstract】 Objective:To observe serum apelin level and its expression in insulin resistance rats which were induced by high-fat feeding,and investigate the relationship between serum apelin level and hyperinsulinemia,glucose and lipid metabolism and body fat parameter.Methods:Total 30 male Wistar rats were randomly divided into two groups,normal control group(NC,n=15) and high-fat-fed group(HF,n=15).Four weeks later,Rats of two groups were performed the hyperinsulinaemic-euglycaemic clamp to confirm that the insulin resistant rats model as duplicated successfully.The level of fasting serum apelin,fasting plasma insulin,fasting plasma glucose,lipid profile,weight and viscera adipose tissue were measured.The apelin expression in rats adipose tissue were tested by RT-PCR.Results:The serum apelin level and expression in adipose tissue in HF was significantly higher than that of NC(p<0.01).Correlation analysis showed that the serum apelin is positively correlated to fasting plasma insulin,fasting plasma glucose,triglyceride,total cholesterol,viscera adipose tissue,weight and free fatty acid(r=0.497,r=0.481,r=0.355,r=0.324,r=0.339,r=0.220,r=0.191 respectively).In a multiple step-wise regression analysis,plasma insulin level is independent facter related to apelin.Conclusion:Serum apelin levels and expression in adipose tissue are increased in insulin resistance rats induced by high-fat feeding.Apelin is correlated with plasma insulin level,glucose and lipid metabolism and body fat parameter.Elevated apelin may contribute to the pathophysiology insulin resistance syndrome.
