亚硒酸钠对实验性自身免疫性甲状腺炎大鼠甲状腺细胞凋亡的影响

【摘要】 目的:探讨亚硒酸钠对实验性自身免疫性甲状腺炎(EAT)大鼠甲状腺细胞凋亡的影响。方法:Wistar大鼠分为正常对照组(A组)、自身免疫性甲状腺炎(EAT)组(B组)和硒干预EAT组(C组)。A组大鼠饮用过滤后的自来水,B、C组大鼠饮用含0.05%碘化钠的水。C组大鼠从实验第2周开始给予亚硒酸钠灌胃,剂量为0.2mg/kg。实验第15天,将猪甲状腺球蛋白(PTg)100mg溶解于灭菌双蒸水50ml中,取10ml与等体积完全弗氏佐剂充分混合成油包水状。取0.5ml(100μg)给B、C组大鼠右后足垫进行皮下注射作为初次免疫。实验第5周,取PTg 10ml(200μg/ml)与不完全弗氏佐剂10ml充分混合成油包水状,取0.5ml(100μg)给B、C组大鼠背部多点皮下注射进行加强免疫,以后每周加强免疫1次,至第10周末实验结束。实验第2、10周末取血,分离血清保存于-70℃。放射免疫分析法(RIA)检测血清中TgAb、TmAb。实验结束,处死大鼠立即取甲状腺,10%中性福尔马林固定,常规石蜡包埋切片HE染色,观察甲状腺形态学变化和淋巴细胞浸润情况。TUNEL法标记甲状腺组织中凋亡细胞。荧光显微镜下观察,细胞核中有黄绿色颗粒者为凋亡细胞。结果:正常对照组、EAT组、硒干预EAT组的TgAb分别(6.94±1.13)、(36.24±3.64)、(17.23±2.90),TmAb分别为(5.96±1.40)、(27.12±5.06)、(15.98±2.45)。硒干预EAT组TgAb、TmAb水平较EAT组明显下降(P<0.05)。HE染色显示,正常对照组甲状腺滤泡呈中度大小,滤泡上皮细胞多呈立方状,滤泡腔内含丰富胶质,小叶周围有少量的血管和纤维,未见淋巴细胞浸润;EAT组表现为甲状腺炎,甲状腺滤泡结构中至重度破坏,淋巴细胞浸润较明显;硒干预EAT组总体结构多数恢复正常,有少量淋巴细胞浸润,滤泡破坏程度减轻,间质内少量纤维组织增生。TUNEL法标记凋亡细胞显示,正常对照组凋亡细胞很少;EAT组凋亡细胞明显增多,且在滤泡结构破坏严重的地方,凋亡细胞较多;硒干预EAT组凋亡细胞明显比EAT组减少,滤泡结构多数恢复正常。结论:亚硒酸钠可能通过抑制甲状腺细胞的凋亡来减轻或抑制自身免疫性甲状腺炎。

【Abstract】 Objective:To investigate the effect of sodium selenite on the rats with autoimmune thyroiditis by observing apoptosis in thyroid tissues.Methods:Wistar rats were divided into three groups:normal control,experimental autoimmune throiditis(EAT) group and selenium curatio-EAT group.EAT model was established by high-dose iodine intake and pig thyroglobulin injection.Rats in selenium curatio-EAT group were given a certain amount of sodium selenite(0.2mg/kg) by intragastric administration.The levels of autoantibodies(TgAb and TmAb),changes of histology of thyroid and apoptosis of thyroid cells through TUNEL were observed.Results:At the end of the experiment,the levels of TgAb in normal control,EAT group and selenium curatio-EAT group were(6.94± 1.13)、(36.24±3.64)、(17.23±2.90) respectively,the levels of TmAb were(5.96±1.40)、(27.12±5.06)、(15.98±2.45) respectively.In selenium curatio-EAT group,the levels of autoantibodies became much lower than EAT group(P<0.05),and the lymphocytes in thyroid were fewer and follicles were better.The number of apoptotic thyroid cells were fewer in selenium curatio-EAT group than in EAT group(P<0.05).Conclusion:Sodium selenite can inhibit the production of autoantibodies of EAT rats,protect the thyroid of rats,and its effects may be operated through suppressing the apoptosis of thyroid cells.
