钙调神经磷酸酶在耐力运动大鼠骨骼肌表型转变中的作用

【摘要】 目的:探讨钙调神经磷酸酶(CaN)在耐力运动时骨骼肌表型转变中的作用。方法:雄性SD大鼠(150~180g)进行6周的无坡度跑台训练(28米/分)。训练大鼠第三周末随机分成二组,其中一组皮下注射环孢素(CSA,15mg/公斤体重/日)3周。第六周末取比目鱼肌和趾长伸肌进行分析。结果:耐力运动时大鼠比目鱼肌纤维向慢肌转换,而CSA阻断该转换。耐力运动时大鼠趾长伸肌CaN活性显著增加,胞核活化T细胞核因子增加,但纤维比例无显著改变;CSA则使耐力运动时趾长伸肌肌原纤维蛋白浓度反增加。结论:CaN参与了耐力运动骨骼肌表型的调控,且具有肌肉特异性。
【Abstract】 Objective:to explore The role of calcineurin(CaN) on changes of skeletal muscle phenotype with endurance training.Methods;Male SD rats((150~200g) were exercised for 6 weeks at 28m/min,grade 0.At the end of third week,half exercised rats were treated with Cyclosporin(CSA,15mg/Kg.BW/day) by hypodermic administration for 3 weeks.After experimental periods,muscle soleus(SOL)and extensor digitorum longus(EDL) were removed for analysis.Results:Endurance Exercise produced fast-to-slow fiber type transformation in the SOL,and CSA prevented these changes.CaN activity In EDL and proteins of nuclear factor of activated Tcells increased significantly,but fiber type composition underwent no obvious changes;CSA resulted in an increased myofibril protein content Of EDL with endurance training.Conclusion:CaN affects Skeletal muscle phenotype with endurance training in a muscle-specific fashion.
