高效液相色谱法测定酮通颗粒中总黄酮醇苷的含量

【摘要】 目的:建立测定酮通颗粒中总黄酮醇苷含量的方法。方法:用高效液相色谱法测定酮通颗粒中总黄酮醇苷含量的含量,色谱柱为DiamonsilTM C18(250mm×4.6 mm,5μm),以甲醇-0.4%磷酸(55:45)为流动相,流速为1.0mL·min-1,检测波长为360 nm,柱温为35℃。结果:槲皮素、山柰素、异鼠李素的线性范围分别为5.24~83.84μg·mL-1(r=0.999 9)、4.56~72.96μg·mL-1(r=0.9999)、2.48~39.68μg·mL-1(r=0.999 9),平均回收率分别为99.84%(RSD=1.26%)、99.61%(RSD=1.06%)、99.39%(RSD=1.08%)。结论:所建立的方法准确、可靠,专属性强,可有效控制酮通颗粒的质量。

【Abstract】 OBJECTIVE To establish a method for determination of total flavonol glycosides in Tongtong granules by HPLC.METHODS The content of total flavonol glycosides in Tongtong granules were determined by HPLC. Chromatographic conditions were as follows:DiamonsilTM C18 column(250mm×4.6 mm,5μm) as the stationary phase,methanol-0.4%phosphoric acid(55:45) as the mobile phase,the velocity of flow was 1.0 mL·min-1 the temperature of coloum was 35℃and the detection wavelength was 360 nm.RESULTS The linear ranges of quercetin,kaempferide and isorhamnetin were 5.24-83.84μg·mL-1(r =0.9999)、4.56-72.96μg·mL-1(r =0.9999)、2.48-39.68μg·mL-1(r =0.9999), respectively.The average recovery were 99.84%(RSD =1.26%),99.61%(RSD =1.06%)、99.39%(RSD =1.08%) respectively.CONCLUTION The method was accurate,reliable and strong specificity.,which can be used to contral the quality of Gengmaitong granules.
