高效液相色谱法测定田银通脉颗粒中三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量

【摘要】 目的:建立田银通脉颗粒中三七总皂苷中三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量测定方法。方法:采用高效液相色谱法测定田银通脉颗粒中三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1的含量。色谱柱为DiamonsilTM C18(250 mm×4.6 mm,5μm);流动相:乙腈(A)-0.05%磷酸水(B)梯度洗脱(0~8 min,20%A:8~40 min 20%A~30%A;40~60 min,30%A~45%A);流速为1.0mL·min-1;柱温为室温;检测波长:203 nm。结果:三七皂苷R1在0.212~3.392μg范围内线性关系良好,r=0.999 6,平均回收率为100.21%,RSD=1.71%(n=9);人参皂苷Rg1在0.832~13.312μg范围内线性关系良好,r=0.999 5,平均回收率为100.24%,RSD=1.62%(n=9),人参皂苷Rb1在0.816~13.056μg范围内线性关系良好,r=0.999 7,平均回收率为99.66%,RSD=1.59%(n=9)。结论:所建立的方法准确、可靠,专属性强,可有效控制田银通脉颗粒的质量。

【Abstract】 OBJECTIVE:To establish a method for determination of notoginsecosideR1,ginsenosideRg1 and Rb1 in Tianyintongmai granules by HPLC. METHODS:The content of notoginsecosideR1,ginsenosideRg1 and Rb1 in Tianyintongmai granules were determined by HPLC.Chromatographic conditions were as follows:Diamonsil? C18 column(250 mm×4.6 mm,5μrn) as the stationary phase,acetonitrile(A)- 0.05%phosphoric acid(B)(0—8 min,20%A;8—40 min 20%A30%A;40—60 min,30%A—45%A) as the gradient mobile phase,the velocity of flow was 1.0 mL·min-1,the temperature of coloum was room temperature and the detection wavelength was 203 nm.RESULTS:The linear ranges of notoginsecosideR1,ginsenosideRg1 and Rb1 fell within 0.212~3.392μg(r =0.999 6),0.832-13.312μg(r =0.999 5),0.816~13.056 ug(r =0.999 7),respectively. The average recovery were 100.21%(RSD =1.71%,n = 9),100.24%(RSD = 1.62%,n = 9),99.66%USD =1.59%,n = 9),respectively.CONCLUTION: The method was accurate,reliable and strong specificity.,which can be used to contral the quality of Tianyintongmai granules.
