Electroacupuncture Alleviated the Motor Symptoms of Unilateral 6-OHDA Mice

【摘要】 Objective:In order to match with our following experiments of optogenetic transgenic mice,we set up a stable unilateral 6-OHDA mice and verify the treatment of electroacupuncture(EA)on the motor symptoms of unilateral 6-OHDA mice.Method:In order to establish a appropriate unilateral 6-OHDA mice model,four dosage(4μg,6μg,8μg and 10μg)of 6-OHDA were injected in right striatum,and the saline was injected as sham group.The mice weight and behavior test,including apomorphine(APO)induced rotation,rotarod test,open field test,were observed for 5 weeks.Then the loss of dopaminergic neurons were determined by the tyrosine hydroxylase(TH)staining with immunohistochemistry.6-OHDA mice administered with 100Hz EA,and the 0Hz was considered as control.Two stainless-steel needled were inserted into the mice at the acupuncture point ST36(ZHUSANLI)and SP6(SANYINJIAO).1.0-1.4 m A electronic stimulation was given for 2 weeks,30min each day and 6 days per week.Results:Compared with sham group,unilateral 6-OHDA lesion induced obvious weight loss and abnormal rotation to the unlesioned side.The statistical results showed that the symptoms of 8μg and 10μg group are more serious.Besides,only 6-OHDA mice in the 8ug group showed a significantly less time on the rotarod,but not in other groups.Meanwhile,both the TH positive neuron and fibers in striatum of lesioned side in 8μg and 10μg group lost more than others.Due to the high mortality rate of 10μg group,the dosage of 8ug6-OHDA was selected to the following experiment.Compared with the 0Hz group,the treatment of 100Hz EA alleviated 8ug 6-OHDA induced motor dysfunction evidenced by increased the time on the rotarod,and horizontal movement distance,movement times and velocity in the open.field test.Conclusion:Injection of 8μg 6-OHDA in striatum could establish a stable Parkinson mice model.Meanwhile the 100Hz EA stimulation was effective in attenuating the movement disorders of unilateral 6-OHDA lesion mice.
