大通湖两种不同壳色形态蚬的分类与系统发育

【摘要】 本研究从传统形态学特征和线粒体COI基因序列信息两方面对大通湖两种不同内壳色(白色和紫色)蚬(n=60)的分类与系统发育关系进行研究。结果显示两种不同壳色蚬的壳高/壳长、壳宽/壳长及壳宽/壳高比值之间无显著差异(P>0.05)。分析样品中发现4种线粒体COI单倍体型,且DT-1(n=38)、DT-7(n=16)和DT-19(n=5)在两种壳色蚬中均有分布,而DT-31仅在一例白色蚬中出现。系统发育树中DT-7与其他3种单倍体型为姐妹枝,且基因序列信息与日本的Corbicula leana完全一致。研究结果表明两种不同壳色蚬在所分析的形态特征及线粒体COI基因序列信息方面无显著差异,故更多线粒体基因与核基因信息以及生理与繁殖等生物学信息将有助于进一步揭示不同壳色形态蚬的分类及亲缘关系。

【Abstract】 Despite its ecological and economic impacts on invasive ecosystems have been one of intensive research topics in freshwater ecology,the taxonomy and systematic status of the genus Corbicula is still unresolved.The argument whether the differences between white and purple were derived from their genetic or environment condition has not come to an agreement.White(n=49) and purple(n=11) morphs of Corbicula clam sympatric in Datong lake were compared using morphological methods(Shell length – SL,Shell hight – SH and Shell width – SW) and diagnostic marker(mitochondrial COI gene) to explore their relationship.Morphological analyses showed no significant differences between the two color morphs for three ratios of SH/SL,SW/SL,and SW/SH either(P> 0.05).Four haplotypes were discovered,and three haplotypes(DT-1,DT-7,DT-19) each shared in both white and purple,the haplotype DT-31 was found only one and within the white morph.Phylogenetic trees,which constructed by NJ and MP of the COI dataset,demonstrated that the four haplotypes were into two divergent clades.Haplotype DT-7 was a sister taxon to the other three,meanwhile suggesting that DT-7 was refer to C.leana due to the identical sequence of COI,the other three still remained uncertain.The results indicated that there was no significant morphological and mitochondrial COI differences among the two color morphs,then a comprehensive study by cross-referencing the mitochondrial-based with nuclear gene data and biological information will help for clarify the taxonomy of the two color morphs in further studies.
