三种消毒剂对草鱼柱状黄杆菌的抑菌效果及最优组方

【摘要】 为了寻找防控草鱼烂鳃病的有效水体消毒剂,首先采用平板划线分离法从患典型烂鳃病草鱼病变部位提取病原菌,根据其形态学特征及生理生化特性对其进行鉴定。然后采用光密度法测定三种水体消毒剂(A、苯扎溴铵,B、戊二醛,C、聚维酮碘)对病原菌的抑菌率,通过正交实验确定三种水体消毒剂抑制病原菌的最优组方。结果显示,该菌株为革兰氏阴性杆菌,平板上菌落黄色,呈树枝状,过氧化氢酶反应阳性,不水解酪氨酸,不产生吲哚,不能发酵和利用葡萄糖,鉴定为柱状黄杆菌。单一用药时,聚维酮碘抑菌效果最好,48 h抑菌率达65%;消毒剂组合抑菌效果最好的为A3B1C2,即苯扎溴铵4.41 g/m3,戊二醛0 g/m3,聚维酮碘100 g/m3,抑菌率达76%。研究表明,聚维酮碘是草鱼烂鳃病的有效水体消毒剂,与苯扎溴铵配伍能增强其抑菌效果。
【Abstract】 To searching for efficacious disinfectant against gill rot disease of Ctenopharyngodon idella,firstly,streak plate method was used to separate the pathogenic strains from diseased regions of Ctenopharyngodon idella with gill rot.Based on analyses of the phenotype,physiologic and biochemical specificity,the pathogenic strain was identified.Secondly,antibacterial effect of three disinfectant(A benzalkonium bromide,B glutaraldehyde,C providone iodine) were determined by densitometry method.Best combination of the three disinfectants was determined by orthogonal experiment.Results showed that the isolated strain was Gram negative bacilli,the colonies on the agar plate showed yellow in the form of a branching mark.Physiologic and biochemical tests showed that catalase tests were positive,the strain couldn’t hydrolyze tyrosine,form indole and ferment glucose.It was identified as Flavobacterium columnare.Using single disinfectant,antibacterial effect of the providone iodine was best and the inhibition reached 65 % in 48 h.Best combination of the three disinfectants was A3B1C2(benzalkonium bromide 4.41 g/m3,glutaraldehyde 0 g/ m3 and providone iodine 100 g/m3),the antibacterial effect was up to 76 %.Our research suggests that providone iodine is the efficacious disinfectant against gill rot disease of Ctenopharyngodon idella,and benzalkonium bromide can raise the sterilization effect.
