鱿鱼墨黑色素及黑色素铁定量分析

【摘要】 采用高速离心法从北太平洋鱿鱼(Ommastrephes bartrami)墨囊中提取黑色素,并通过黑色素与FeCl3螯合制备了Fe(III)含量为7%(W/W)的黑色素铁。采用H2O2氧化降解及高效液相色谱法(HPLC)定量测定黑色素两种降解产物2,3-二羧酸吡咯(PDCA)和2,3,5-三羧酸吡咯(PTCA)对鱿鱼墨黑色素及黑色素铁进行了定量研究。结果表明:以H2O2氧化降解黑色素产物PDCA和PTCA为基础的鱿鱼墨黑色素定量方法效果良好。利用该方法对黑色素铁进行定量仅检测出40%的黑色素,盐酸脱铁后检出量上升至75%。常规的鱿鱼墨黑色素的定量方法不适用于黑色素铁的定量分析。
【Abstract】 Extracted melanin from ink of North Pacific squid (Ommastrephes bartrami) by high-speed centrifugation. Prepared squid melanin-Fe (Ⅲ) with the method of exposing squid melanin to aqueous solution of FeCl 3 , the content of bound Fe (Ⅲ) was 7%(w/w) of melanin. Oxidative degradation with H 2 O 2 and quantify squid melanin with HPLC by determine two degradation products pyrrole-2,3-dicarboxylic acid (PDCA) and pyrrole-2,3,5-tricarboxylic acid (PTCA) of melanin. Results showed that the quantitative method of squid melanin that based on two degradation products PTCA and PDCA of melanin by H 2 O 2 had good result. With this method to quantify squid melanin-Fe (Ⅲ), the melanin detected was only 40%. After hydrochloric acid treatment to expel Fe, the melanin detected rised to 75%.The conventional quantitative method of squid melanin was not used to quantify melanin-Fe (Ⅲ).
