鲫鱼肠道微生物区系及其来源分析

【摘要】 肠道细菌群落影响宿主的生长,食物转化,上皮发育,免疫以及病原微生物在肠道的建立。因此,肠道微生物受到了越来越多的关注。本研究用焦磷酸测序(pyrosequencing)的方法详细研究了鲫(Carassius auratus gibelio)消化道微生物区系及其来源。研究结果显示鲫消化道和养殖环境栖息着复杂的微生物类群,其中变形菌门(Proteobacteria)是最主要的类群,然后是厚壁菌门(Firmicutes)和放线菌门(Actinobacteria),而拟杆菌门(Bacteroidetes)只占很小的比例。潜在的益生菌(乳酸杆菌属Lactobacillus,乳球菌属Lactococcus和芽孢杆菌属Bacillus)和病原菌(气单胞菌属Aeromonas和不动杆菌属Acinetobacter)在肠道中低丰度但稳定存在。细菌群落比较分析显示鲫肠道菌群与底泥微生物群落最相似,表明底泥细菌群落是影响鲫肠道菌群的最重要的因子。然而,饲料中37.95%OTUs出现的肠道中,说明食物中的细菌明显的影响鲫肠道微生物区系,益生菌可以添加到饲料中。
【Abstract】 In this study,pyrosequencing has been used to explore the characterization of the microflora of intestine of gibel carp(Carassius auratus gibelio) and the origin of these intestinal microorganisms.The results disclose bacterial diversity in the cultured environments and the fish intestinal content is unexpectedly high.The gibel carp harbors characteristic intestinal microflora, where Proteobacteria is predominant,followed by Firmicutes,Actinobacteria,while Bacteroidetes is relatively low abundant.The potential probiotics(Lactobacillus,Lactococcus and Bacillus) and pathogens(Aeromonas and Acinetobacter) are of low abundance in the intestinal microbiota. Bacterial community comparisons show that communities of JYN(JYNC,JYNa and JYNb) is most closely related to that of the sediment community,indicating that microbiota of the sediment is the most important factor in determination the bacterial community of gibel carp gut.However, 37.95%OTUs presented in PHSL community occur in JYN communities,suggesting that bacteria in food play remarkable influence on the microflora of gibel carp intestine,and that probiotics may be supplemented in feed.
