添加微生态制剂对主养草鱼水体中细菌群落的影响

【摘要】 有关益生菌对养殖水质的改良作用及其机理至今尚未明确。本实验中检验了添加3种商业微生态制剂(诺碧清生物净水剂、光合细菌、EM菌)对主养草鱼水体细菌群落的影响。实验设4个处理(对照、诺碧清生物净水剂、光合细菌、EM菌),每个处理设3个重复。实验容器为2000L聚乙烯水槽,每个水槽中放养25尾草鱼、5尾鲫鱼、2尾鲢鱼。实验期间每天向水槽投喂鱼配合饲料,定期采样分析溶氧、pH、氨氮、硝酸盐、亚硝酸盐、磷酸盐、化学耗氧量、细菌生物量和细菌种类组成。细菌种类根据变性梯度凝胶电泳(DGGE)和DNA测序方法确定。实验结果显示:各水槽内细菌种类组成随时间推移变化明显,添加不同微生态制剂的水槽内细菌种类组成存在明显差异。仅有少数加入的益生菌种类在实验水槽内被检测到,这意味着部分益生菌可以在水中定殖。加入3种复合益生菌制剂均未导致上述水质指标发生显著变化,细菌生物量与上述水质指标均无显著相关.
【Abstract】 The effects of adding commercial microecologics on bacterial community in waters for grass carp farming were examined.Four treatments,including control,Novozymes pond plus, photosynthetic bacteria and EM bacteria were established.The experiment was conducted in 2000 L plastic tanks,in each tank 25 grass carp,5 crucian carp and 2 silver carp were stocked and fed formulated fish feed.During the experiment,dissolved oxygen,pH,ammonia,nitrate,nitrite, reactive phosphate,chemical oxygen demond,species and biomass of bacteria in the tankes were determined.The bacteria species was identified by denaturing gradient gel electrophoresis(DGGE) and DNA sequence analysis.The bacterial species composition in each tank changed considerably at different stage of the experiment,and significant difference was detected in bacterial species composition among the treatments.Only few added bacteria were detected in the tanks,suggesting these bacteria could settled in native bacterial communities.No significant differences were found in water quality parameters among the treatments,and no significant correlation was detected between bacteria biomass and water quality parameters.
